摘要:Fe3+、Zn2+、Cu2+等过渡金属离子对于环境的影响很大,同时也在生物体内扮演着十分重要的角色,所以对这些离子的分析检测具有重大意义。罗丹明系列衍生物具有良好的光物理性质,如摩尔消光系数大、发射波长长、荧光量子产率高。因此被广泛使用在荧光分子探针的设计合成中。本文以罗丹明B,水合肼、对羟基苯甲醛为基础原料,由取代、亲核加成等一系列反应合成了一种新型的分子探针H1,经1H-NMR、质谱对探针H1进行结构表征后,确认产物与目标化合物结构一致,产率为40.7%。同时通过荧光光度计检测其性能后发现,该探针H1在pH为7.02的CH3CN/H2O (95/5,V/V)溶液中可高选择性识别Fe3+,常见的大部分的金属阳离子对Fe3+识别测试无明显干扰。当加入Fe3+后,探针H1的内酰胺环结构发生开环,在λem=582.5 nm附近处荧光明显增强,且溶液颜色由无色转变为桃红色,实现了裸眼识别的效果。最后测试表明探针H1和Fe3+形成了 1:1的络合物,当络合后的溶液再加入20倍的EDTA后,溶液荧光强度急剧下降,说明这是一个可逆的识别过程。32051

The studies on synthesis of rhodamine-based fluorescent probe and properties of recongnition for Fe3+
Abstract: Some heavy metal ions play important roles in environment and living systems, such as Fe3+, Zn2+, Cu2+ etc. Thus the analysis and detection of those ions has a great significance. Rhodamine derivatives are highly favorable because of their excellent photopysical properties, such as high extinction co-efficients, excellent quantum yields, great photostability and relatively long emission wavelengths. In this paper, rhodamine B, hydrazine hydrate and p-hydroxy benzaldehyde as raw materials were used to synthesize a new type of molecular probe H1, whose yield was 40.7%, through substitution reaction, nucleophilic addition reaction and other reactions. By 1H-NMR, mass spectrometry, compound structure was consistent with the target product. At the same time, by fluorophotometer, probe H1 was proved that the probe H1 was selectively detected iron ions in CH3CN/H2O (95/5,V/V) at pH 7.02, and most of metal ions on the determination of Fe3+ have no significant interference. Otherwise, with the increase of Fe3+ concentration, fluorescence was enhanced obviously at λem=582.5 nm. And the color of solution changed from colorless to pink, which can be recognized by naked eyes in practical application. Finally, the ring of lactam of probe H1 may open and probe H1 and Fe3+ may form a complex (1:1). After adding 20-fold EDTA, the solution fluorescence intensity fell sharply, which showed that an identification of the reversible process occurred.
