银纳米颗粒对血液荧光增强效果的时间作用研究

摘要:本文采用FLS900型稳态和瞬态荧光光谱仪进行了银纳米颗粒对血液荧光增强效果的时间作用研究。从实验上研究了全血溶液和加入银胶的全血溶液荧光光谱随时间变化情况,并分析了其光谱各个强度峰值处的荧光强度以及荧光增强倍数随时间的变化,得出结论:银纳米颗粒能与血液中的黄素腺嘌呤二核苷酸、血卟啉环、锌卟啉发生荧光增强效应,分别对应荧光光谱309nm,560nm和593nm处;在样品配制完成后2小时以内均出现荧光增强现象;3小时以后增强效果明显降低;黄素腺嘌呤二核苷酸在样品配制完成后2小时荧光增强效果最佳,血卟啉环,锌卟啉在配制完成时荧光增强倍数最大。通过分析产生荧光增强效应的机理得出其原因可能是银纳米粒子和稀释全血溶液中的荧光物质在配制完成后的不同时间内距离不一样导致产生荧光增强的效应强弱不同。本文的研究结果对血液中黄素腺嘌呤二核苷酸、血卟啉环及锌卟啉的荧光光谱的研究及其在疾病临床诊断上的应用具有一定意义,并且为时间因素对其他荧光光谱影响的研究提供了案例。38362

Study on Time Effect of Silver Nanoparticles on the Fluorescence Enhancement of Bloods
Abstract: Time effect of silver nanoparticles on the fluorescence enhancement of bloods was investigated by the FLS900 steady-state and transient fluorescence spectrometer in this paper. The fluorescence spectrum of whole blood solution and whole blood of silver sol solution are investigated experimentally by time,and the fluorescence intensity and fluorescence enhancement ratio of each fluorescence emission peak in the spectrum are analyzed theoretically. The result showed:in the blood of the silver nanoparticles occurs fluorescence enhancement effect with flavin adenine dinucleotide, blood porphyrin rings, and zinc porphyrin ,in wavelength of  309 nm, 560 nm and 593 nm in the fluorescence spectrum; fluorescence enhancement phenomenon occurs within 2 hours after completion of the sample preparation but 3 hours after ,it significantly reduced; the fluorescence enhancement effect of Flavin adenine dinucleotide is best 2 hours later after sample preparation, blood porphyrin rings, zinc porphyrin achieve the biggest fluorescence enhancement ratio just after the completion of preparation. After discussing the mechanism of the fluorescence enhancement effect, it is considered that the different distance between silver nanoparticles and fluorescent material in whole blood solution in each period after completion of the preparation influence the strength of fluorescence enhancement effect. The research in this paper can offer reference to the study of fluorescence spectrum about flavin adenine dinucleotide, porphyrin rings and zinc porphyrin and their application in clinical diagnosis, and also provides the case for research in other fluorescence spectrum which discusses time effect.
