利用光阱力测量血红细胞膜的弹性模量

摘要光镊技术是生物学、物理学和医学的研究热点中的一项前沿技术,基于光学微操纵技术,具有无损伤、非接触等优点。首先采用光阱力对血红细胞和二氧化硅微球进行捕获,通过设计光阱的行进路径,拉动吸附在血红细胞上的二氧化硅微球,记录运动轨迹及光阱力的大小。利用胡克定律和牛顿第二定律可以算出血红细胞膜的弹性模量。采用同样的实验方法测量了糖尿病人的血红细胞,得到病变血红细胞的膜弹性物理参数, 通过比较正常和病变血红细胞膜剪切弹性模量的不同,结合细胞膜理论进行了机理分析,认为是病变改变了血红细胞膜的组成成分导致了这种差异。 正常红细胞和病变红细胞膜弹性的不同的结果可以对探索血红细胞膜的结构功能,评价血液质量,分析病变原因等提供有价值的参考,相应的研究对疾病的药物治疗效果也具有重要的应用价值。该论文有图13 幅,表4 个,参考文献17 篇。51403 毕业论文关键词:光镊 血红细胞膜 光阱力 弹性模量 Measuring the Elastic Modulus of Red Blood CellMembrane by Optical Trapping Force Abstract Optical tweezers technology is a frontier technology in the research of biology,physics and medicine.It is based on optical micro manipulation technology and is nodamaged, non-contacted.Firstly, the red blood cells and silica microspheres werecaptured by optical trap force,through the path of a moving optical trap,pulling thesilica microspheres adsorbed on the blood red blood cells,recording the motiontrajectory and the size of the optical trapping force.Hooke's law and Newton's secondlaw can be used to calculate the elastic modulus of the red blood cell membrane.Thesame experimental method was used to measure the blood red blood cells in thepatients with diabetes mellitus.The membrane elastic physical parameters of thediseased blood red blood cells were obtained.By comparing the shear elastic modulusof normal and diseased blood red blood cell membranes,combined with the cellmembrane theory we analyzes the mechanism.We think the difference is caused bythe change of the blood red cell membrane.Different results of membrane elasticity ofnormal red blood cells and diseased red blood cells can explore the structure andfunction of erythrocyte membrane,evaluate blood quality,analyses the causes of thedisease to provide a valuable reference.The corresponding research has importantapplication value for the drug treatment of the disease.
