加拿大一枝黄花组织总黄酮的提取和抗氧化研究

摘要用超声萃取法提取加拿大一枝黄花叶片组织中的总黄酮成分,以比色法测定其 含量,同时比较总黄酮的季节变化。以 Vc 做对照,对加拿大一枝黄花叶片黄酮浸 提液进行自由基清除及抗氧化活性试验。采取 AB-8、D101 和 NKA-Ⅱ三种不同类 型的大孔吸附树脂对黄酮浸提液进行静态吸附实验与解吸实验。实验结果显示,加 拿大一枝黄花叶片中各月份蛋白质和总黄酮含量均呈现先增后减趋势,叶片中蛋白 质的含量在 2 mg/g~5 mg/g 之间,以 7 月份含量为最高,大约在 5 mg/g。叶片中总黄酮的含量约在 50 mg/g~300 mg/g 之间,以 6 月份含量为最高,大约在 300 mg/g; 提取物清除自由基及抗氧化活性的能力与总黄酮的含量呈线性递增关系,其中清除 过氧化氢和抗脂质过氧化能力强于同浓度的 Vc,而清除 DPPH、羟基自由基能力及 还原能力则弱于 Vc。静态吸附与解吸实验表明,AB-8 型大孔吸附树脂对组织总黄 酮的吸附率达到 64。15%,解吸率达 33。14%,总黄酮回收率达 21。26%;结合静态吸 附和解吸动力学曲线,发现 NKA-Ⅱ型树脂对加拿大一枝黄花组织黄酮浸提液表现 出了较好的吸附性能,而 D101 型树脂对加拿大一枝黄花黄酮浸提液表现出较好的 解吸性能。80721 毕业论文关键词:加拿大一枝黄花;总黄酮;抗氧化活性;大孔吸附树脂;静态吸附与解吸 Abstract In this paper, the content of total flavonoids extracted from leaves of Solidago canadensis L。 by ultrasonic method and the seasonal change of total flavonoids were studied。 Using VC as control, the experiments of free radical scavenging and antioxidant activity were carried out on the total flavonoids extract。 And 3 different types of resins, AB-8、D101 and NKA-Ⅱ, were used for purifying the total flavonoids extract with the static adsorption and desorption experiments。 The results showed that the contents of the proteins and total flavonoids from Solidago canadensis L。 were different from April to November。 The contents of the proteins increased from 2mg/g to 5mg/g and the contents of total flavonoids increased from 50 mg/g to 300 mg/g。 The highest contents of proteins and total flavonoids were reached in July and June respectively。 The free radical scavenging and antioxidant activity of flavonoids extract have a dose-effect relationship with the content。 The static adsorption and desorption experiments reveal that, adsorption ratio of AB-8 resins and desorption ratio reach to 64。15% and 33。14%, respectively; the total flavonoids recovery ratio reaches to 21。26%。 The static adsorption and desorption kinetics curve display that, NKA-Ⅱresins perform better in adsorption and D101 resins perform better in desorption。 Keywords: Solidago canadensis L。; total flavonoids; antioxidative activity; macroporous resin; static adsorption and desorption 目 录 第一章 绪论 1 1。1 一枝黄花属植物简介 1 1。2 加拿大一枝黄花的危害 1 1。2。1 对本土植物的危害 2 1。2。2 对农业生产的危害 3 1。3 加拿大一枝黄花的生长和繁殖特性 3 1。4 加拿大一枝黄花的防治措施 4 1。5 黄酮类化合物研究简介 5 1。5。1 酮类化合物的来源 5 1。5。2 黄酮类化合物的药理活性 (责任编辑:qin) |
