摘要:为明确细胞色素P450在绿盲蝽毒死蜱抗性中的作用,本文通过转录组测序技术分析了绿盲蝽毒死蜱抗性品系中细胞色素P450超基因家族的表达水平,同时利用实时定量PCR方法验证了相应P450基因的相对表达量。转录组数据分析表明:在绿盲蝽体内共有75个P450基因,与敏感品系SLF和BZS相比,毒死蜱抗性品系BZR有6个P450基因表达量显著上升,分别为CYP6A20、CYP6A14、CYP6X1v1、CYP6B6、CYP6BS1、以及CYP4ac1。而实时定量PCR验证的结果则表明在BZR品系中仅CYP6B6、CYP6BS1、CYP6a20和CYP6X1v1基因的表达量明显提高,分别为SLF品系中的9.21、7.38、7.90和2.86倍,差异达到显著水平。因此可以推断,CYP6B6、CYP6BS1、CYP6a20和CYP6X1v1可能参与了绿盲蝽BZR品系毒死蜱抗性的形成。26628 毕业论文关键词:绿盲蝽;P450基因;毒死蜱;抗性
The relative expression of P450 genes in the resistant strain to chlorpyrifos of Apolygus lucorum
Abstract:Relative expression of P450 genes in chlorpyrifos-resistant strain (BZR) was analyzed using transcriptome sequencing and confirmed by quantitative real-time PCR to indentify the role of P450 genes in the resistance of Apolygus lucorum to chlorpyrifos. Transcriptome sequencing data showed that the relative expression level of 6 among 75 P450 genes in BZR were up-regulated. These genes were CYP6A20, CYP6A14, CYP6X1v1, CYP6B6, CYP6BS1 and CYP4ac1. QrtPCR on these genes showed only CYP6A20, CYP6X1v1, CYP6B6, CYP6BS1 were significantly higher expressed. The expression of these genes in BZR was 9.21-, 7.38-,7.90- and 2.86-fold higher than that in susceptible stain SLF. These results indicated CYP6A20, CYP6X1v1,CYP6B6, CYP6BS1 might be involved in the resistance to chlorpyrifos of A. lucorum.
