摘要:Microbulbifer sp. A4B-17菌株拥有合成对羟基苯甲酸(4-Hydroxybenzoic acid,4HBA)及其酯类的功能。4HBA在工业生产和日常生活当中有很多重要的作用,例如:在染料工业中用于合成热敏染料的显色剂;可用于彩色胶片及合成油溶性成色剂“538”及尼龙12中用作增塑剂的生产原料;还可用于液晶聚合物和塑料。本研究旨在探讨A4B-17菌株合成4HBA及其酯类的最佳碳源,为将A4B-18菌株开发成高效工业菌株奠定基础,从而使生产过程更加清洁,环保。实验结果如下:(1)在无机海水培养基(ONR7a)中分别加入各种糖类作为唯一碳源对A4B-17进行培养,发现除了丙酮酸外其他碳源的长势都不是很好,可能是因为该菌株需要生长因子的原因。(2)为了对4HBA进行定量分析,配制不同浓度的4HBA进行高效液相色谱分析绘制了标准曲线,然后将A4B-17菌株在4种不同的培养基中培养,对合成的4HBA定量分析。结果表明MB培养基,海水营养培养基,葡萄糖培养基和丙酮酸培养基中分别合成了0.057mMol/L, 0.007mMol/L,0.00059mMol/L和0.00068mMol/L的4HBA。39891 毕业论文关键词:对羟基苯甲酸;高效液相色谱;生物合成
Study on the synthesis of 4-hydroxybenzoic acid by Microbulbifer strain sp.A4B-17
Abstract: Microbulbifer sp. A4B-17 strain with synthesis of p-hydroxybenzoic acid (4-Hydroxybenzoic, acid, 4HBA) and the ester function.4HBA in industrial production and daily life has a lot of important role, such as in industrial dye for synthesis of heat sensitive dye chromogenic agent; can be used for color film and synthetic oil soluble couplers 538 and nylon 12 as plasticizing agent in the production of raw materials; also can be used for liquid crystal polymers and plastics.This study designed to explore the best carbon source for the A4B-17 strain synthesis 4HBA and its esters, for A4B-18 strains developed into efficient industrial strains to lay the foundation, so as to make the production process more clean and environmentally friendly.The experimental results are as follows: (1) in seawater inorganic training base (ONR7a) were added sugars as sole carbon source of A4B-17 were cultured, found in addition to pyruvate other carbon sources of growth are not very good, may because of the strain required growth factor.(2) in order to make a quantitative analysis on the 4HBA prepared different concentrations of 4HBA performed by high performance liquid chromatography analysis draw the standard curve, then A4B-17 strains in 4 kinds of different culture media culture, and analyses on the synthesis of 4HBA quantitative. The results show that the MB medium, seawater nutrient medium, glucose culture medium and pyruvate culture medium were synthesized0.057mMol/L,0.007mMol/L, and 0.00059mMol/L 0.00068mMol/L 4HBA.
