摘要本实验利用聚合酶链式反应-单链构象多态性(PCR-SSCP)和DNA测序技术,研究湖羊TGFBR2基因的遗传变异,来探讨该基因的遗传多态性与湖羊体尺性状(体长、体高、胸围、管围等)的关系,以期发现对其体尺性状具有显著效应的遗传标记(genetic marker),为湖羊的进一步选育提供理论依据。实验共采集了80只湖羊耳组织样本,分别提取总DNA,纯化后进行PCR扩增,对扩增情况好的基因进行PCR-SSCP检测,多态产物纯化测序后进行序列分析以及基因型的判定,分析遗传多样性、遗传特征及其与湖羊的体尺性状进行相关性。结果显示:在湖羊TGFBR2基因上共检测到一个多态位点,2种基因型(AA和AB);A基因频率为0.85,为优势基因;多态信息含量(PIC)为0.192,为低度多态;该位点与体长、体高、胸围、管围的相关性均为不显著(P>0.05),说明该突变位点对湖羊体尺性状进行选择的余地较小。71680

Preliminary analysis of genetic persity of TGFBR2 Gene in Hu sheep
Abstract In this study, using the polymerase chain reaction-single strand conformation polymorphism (PCR-SSCP) and DNA sequencing techniques to study genetic variation of TGFBR2 gene of 80 Hu sheep .To investigate the relationship between genetic polymorphism and body size traits (body length, body height, girth, cannon circumference, etc.) of Hu sheep, in order to find the economically important traits with significant genetic markers and provide theoretical basis for further breeding. Firstly, collecting 80 Hu sheep’s ear tissues, then extracting total genomic DNA, performing PCR amplification, followed by SSCP test. Polymorphic loci will be sequenced to determine the mutation site, and make correlation analysis with sheep’s body size traits. The results showed that: two genotypes (AA and AB) and one polymorphic loci were detected on TGFBR2 gene of Hu sheep; A Gene is the dominant gene and the frequency of it is 0.85; polymorphic information content (PIC) is 0.192 , which is low polymorphism; Different genotypes mutation on body length, body height, girth, cannon circumference correlation is not significant (P> 0.05),which indicating that there is little space to select body size traits of Hu Sheep at this point.
