摘要本实验采用Real-Time PCR 的方法,研究分析TSC1基因在徐淮山羊胎羊的不同组织(心、肝、脾、肺、肾、肠、背肌、腿肌)间的表达差异性,为研究山羊TSC1基因的表达规律奠定基础。实验过程为首先采集徐淮山羊胎羊的心、肝、脾、肺、肾、肠、背肌、腿肌等部位的组织,提取出八个组织中的总的RNA,测量并记录RNA的浓度和纯度。再将RNA反转录得到cDNA。最后用聚合酶链式反应(PCR)方法处理cDNA检测出TSC1基因在徐淮山羊胎羊的八个不同组织中的表达水平。实验显示,TSC1基因在八个组织中均有一定量的表达,但表达水平各不相同,比如在心肌中的表达量远远高于其他组织,而肺中的表达量相对来说则是微乎其微,其他组织中也都有表达;TSC1 在徐淮山羊胎羊的各组织中表达量具体数据为:心肌(1.0000)>腿肌(0.1833)>肝(0.1277)>背肌(0.1236)>肾(0.1194)> 肠(0.0263)>脾(0.0045)>肺(0.0044)。该结果可作为对TSC1基因进一步研究的实验基础和理论依据。71683

The Study on Different Expression of TSC1 Gene in Different Fetal Goat Organs
Abstract In this experiment , we made research and analysis of expression of TSC1 gene in different organizations (heart,liver, spleen,lung,kidney,intestines,back mascle,leg mascle)of xuhuai fetal goat with relative quantification PCR technology for further study on functions of TSC1 in different organs. The first step of the experiment is to collect different organizations of fetal sheep. By means of extracting the total RNA in xuhuai goat eight tissues including heart , liver , spleen , lung , kidney , intestines , back muscle , leg muscle to measure and record the potency and the purity of RNA . Then the RNA would be retroactively transcribed into cDNA with RNA primer. In the last, the experiment used Real-Time quantitative PCR to deal with cDNA, detecting the expression of TSC1 gene in eight tissues of xuhuai fetal goat. In the research, we can found that the TSC1 gene successfully expressed in each tissue , but the quantities of expression of TSC1 varied widely: The highest level of expression was in heart and the lowest expression level was in lung . The specific figures were as follows: heart(1.0000)> leg muscle(0.1833)>liver(0.1277)>back muscle(0.1236)> kidney(0.1194) >intestines(0.0263) > spleen(0.0045) >lung(0.0044). The result could be used in further study of TSC1 as experiment basis and theoretical basis.
