摘要当前在提取工艺优化中,常用的方法为正交试验法与响应面法,但选择何种优化 方法尚无依据。本文在单因素试验结果的基础上,分别采用正交优化法和响应曲面法 就提取温度、乙醇浓度、液料比和提取时间四个因素优化桑叶黄酮的提取工艺,通过 分析两种方法所得的优化结果,明确适合桑叶黄酮提取工艺优化的方法。通过正交试 验法优化得到的桑叶黄酮提取的最佳工艺为:提取温度 60 ℃、提取时间 60 min、乙 醇浓度 50%、液料比 100 mL/g,在此条件下,桑叶黄酮的提取得率为 4。53%;通过响 应面试验优化得到的桑叶黄酮最佳提取工艺为:提取温度 68 ℃、提取时间 40 min、 乙醇浓度 52%、液料比 86 mL/g,此条件下桑叶黄酮的得率为 4。71%,而理论预测值 为 4。64%。结果表明:响应面法优化的工艺条件下得到的桑叶黄酮量高于正交试验法, 生产能耗也低于后者;并且响应面法建立的优化模型也能较好地预测桑叶黄酮的提取 率。研究结果可为类似工艺选择优化方法提供一定的借鉴和指导。80977

Abstract At present, orthogonal design (OD) and response surface methodology (RSM) are commonly used for optimizing extraction process, although there is no specific scientific evidence that which method is optimum。 In this article, with the basis of single factor experiment, four appropriate factors including extraction temperature, ethanol concentration, the ratio of liquor to material (liquor-material) and extraction time were investigated by means of OD and RSM to optimize the extraction process of flavonoids in mulberry leaves。 The optimization results based on two different methods were analyzed, in order to fide which one is suitable for optimizing the extraction process of flavonoids in mulberry leaves。 The extraction conditions optimized with OD for flavonoids was
extraction temperature of 60 ℃, extraction time of 60 min, ethanol concentration of 50%
and liquor - material of 100 mL/g, with a flavonoids yield of 4。53%。 The optimized process from RSM was extraction temperature of 68 ℃, extraction time of 40 min, ethanol
concentration of 52% and liquor - material of 86 mL/g, with a flavonoids yield of 4。71%, while the predicted yield of 4。64%。 The result indicates that the process optimized with RSM can obtain more flavonoids than that of OD, and it consumes less energy than the later; in addition, the model established with RSM has good fitting performance to predict flavonoids yield of mulberry leaves。 The result can provide some valuable reference and guidance for the choice of optimal method in similar process。
