摘 要 目的探讨腺苷酸环化酶3 (adenylate cyclase 3,ADCY3)基因多态性与冠状动脉粥样硬化性心脏病(coronary heart disease,CHD)的关系,并分析其多态性与环境因子是否有交互作用。方法采用病例对照研究方法,随机抽取并调查了浙江省宁波市某区常住居民2279人。筛选了ADCY3基因12个标签单核苷酸多态性位点(tag SNP),运用连接酶检测反应技术检测基因分型。应用多因素logistic回归模型分析ADCY3基因多态性与冠心病的关系,并运用叉生分析判断其基因多态性与环境因子是否可能存在交互作用。结果携rs4665273位点CT基因型者冠心病风险是带CC基因型者的4。13倍(OR=4。13,95%CI=1。13-15。04);携rs6751537位点GG基因型冠心病风险是带AA基因型的54。49倍(OR=54。49,95%CI=6。15-482。72);携rs2278485位点AG基因型冠心病风险是带AA基因型个体的2。40倍(OR=2。40,95%CI=1。18-4。87)。叉生分析显示有10个位点多态性与饮酒交互作用可能和冠心病相关。结论ADCY3基因多态性与冠心病有关联,且饮酒可能对该基因多态性与冠心病的关联存在一定影响。90033
Objective To investigate the relationship between adenylyl cyclase 3 (ADCY3) gene polymorphism and coronary heart disease (CHD) risk, and to analyze whether the interaction between the polymorphism and environmental factors。 Methods By using a Case - control study design, 2279 participants were randomly selected and investigated in Ningbo city Zhejiang Province。12 loci of ADCY3 gene were selected by tag SNP method, and genotypes were detected by ligase detection reaction technique。 Multivariate logistic regression model was used to analyze the relationship between ADCY3 gene polymorphism and CHD, and crossover analysis was used to analyze the potential interaction between the gene polymorphism and environmental factors。 Results CT genotype of rs4665273 was associated with 4。13 times higher risk of CHD (OR=4。13, 95%CI=1。13-15。04;compared with CC genotype); GG genotype of rs6751537 was associated with 54。49 times higher risk of CHD (OR=54。49, 95%CI=6。15-482。72; compared with AA genotype); AG genotype of rs2278485 was associated with 2。40 times higher risk of CHD (OR=2。40, 95%CI=1。18-4。87; compared with AA genotype)。 The results of crossover analysis showed that there may be interaction between gene polymorphism and alcohol。 Conclusion ADCY3 gene polymorphism is associated with CHD, and drinking may have some influence on the association between the gene polymorphism and coronary heart disease。
