摘要:本课题以葛根为原料,利用超声波提取其中的黄酮类物质,并用双水相萃取体系对它进行初步的分离纯化。利用单因素实验和正交实验对超声提取的工艺条件进行优化;通过对比四种双水相体系的分相能力和萃取能力,确定最适的双水相萃取体系,并对萃取体系的最佳纯化条件进行了研究。结果表明:超声辅助提取葛根黄酮类物质的最佳条件为:超声时间30min,超声功率为150W,超声温度为60℃,料液比为1:20。最佳萃取体系为乙醇/磷酸氢二钾双水相体系,该体系得到最佳萃取条件:乙醇质量分数为42.86%、磷酸氢二钾质量分数为26.42%,pH为8。61940

Study on Ultrasonic Assisted Extraction of Flavonoids from Puerarin and Preliminary Purification
Abstract: In this paper Puerarin as raw material, the use of ultrasonic extract of flavonoids and aqueous two-phase extraction system with its preliminary separation and purification. By single factor and orthogonal experiments on ultrasonic extraction process conditions are optimized; the comparison of four kinds of aqueous two-phase system of phase separation and extraction ability to determine the optimal aqueous two-phase extraction system and the best extraction system purification conditions. The results showed that ultrasonic assisted extraction of flavonoids in Pueraria optimal conditions is ultrasonic time of 30min, ultrasonic power of 150W, ultrasonic temperature of 60 ℃, solid-liquid ratio of 1:20. The best system for the extraction is ethanol / dibasic potassium phosphate aqueous two-phase system which is optimum extraction conditions: ethanol mass fraction of 42.86%, dipotassium hydrogen mass fraction of 26.42%, pH 8.
