摘要为了建立高效液相色谱紫外法测定市售烤肉中的3,4-苯并芘,本论文对烤肉的前处理过程进行考察优化,探索出合理的烤肉前处理方法,并对色谱条件进行优化。结果具体如下:正己烷超声波提取30min,含2 mol/L KOH 的甲醇∶水(9∶1;v/v)溶液水浴皂化1h,环己烷液液萃取,浓缩。前处理后进行高效液相紫外法分析,色谱柱Hypersil ODS C18,4.6mm*250mm;柱温25℃;检测波长295 nm;流动相组成甲醇:水 = 95 : 5;流速1.0 mL/min;进样量10 μL。回收率为57.79%到77.26%。回归方程为y = 10.248x + 12.754,R2 = 0.9992。用此方法对烤肉中苯并芘进行检测分析比较快捷方便,同时具有经济性,适合于检测烤肉中3,4-苯并芘。9376

Title Determination of Benzo(a)Pyrene in roast meat by High Perfomance Liquid Chromatography - UV
Abstract To develop a method for determining Benzo(a)Pyrene in roast meat sold in market by HPLC-UV, investigated the proper method of pretreatment and chromatographic conditions.The results were as follows: Extract with normal hexane by Ultrasonic Extraction, saponify with 2mol/L KOH in methanol: water(9:1; v/v) solution, liquid-liquid extraction with cyclohexane, concentrate.Then analyzed by HPLC and the chromatographic conditions were as follows: column:Hypersil ODS C18,4.6mm*250mm,25℃; detection wavelength: 295nm; mobile phase: methanol:water(95:5;v/v);flow rate: 1.0mL/min; injection volume: 10μL.The recoveries were between 57.79% and 77.26%. The linear equation was y = 10.248x + 12.754,R2=0.9992.Because it was quick ,easy and thrifty to determine Benzo(a)Pyrene by this method, it could be the appropriate one.

1 引言
