摘要紫藤系豆科紫藤属,是一种落叶攀援缠绕性大藤本植物,随着人们对紫藤化学成分以及药理作用的相关研究,初步确定其根茎叶中含有生物碱、黄酮类、蛋白质、糖及苷类等多种化学成分,黄酮类化合物具有许多重要的药理作用,能够抑制酶活、抵制癌症、抵制病毒、抗炎、抗过敏、抗糖尿病并发症等作用,没有毒害作用,能够有效治疗肿瘤、衰老、心血管等疾病。实验目的:研究超声波辅助从紫藤叶中提取黄酮类物质的工艺和超声频率、超声时间、物料比、乙醇浓度这四个因素对紫藤叶总黄酮提取量的影响,并通过响应面法优化其提取条件。实验方法与内容:根据芦丁标准曲线回归方程由吸光度求出了各处理一定体积提取液中黄酮的含量,并用单因素试验探究不同超声频率、萃取时间、料液比、乙醇浓度对总黄酮提取量的影响,评价不同因素对紫藤叶总黄酮提取量的影响程度,然后用响应面法确定最佳提取工艺并评价不同因素之间的交互作用。实验结果:用超声法从紫藤叶中提取黄酮类物质的最优条件是超声时间46min、料液比1:48、乙醇体积分数25%,提取量达到了7。35 mg/g,且其提取物能够清除羟自由基和超氧阴离子自由基,清除率在一定范围内随总黄酮质量浓度的增加而增强。75029

Ultrasonic-assisted Extraction of Flavonoids from Wisteria sinensis through Response Surface Optimization to Evaluate the Experiment
Abstract Wisteria sinensis belongs to the leguminous wisteria genera and it is a kind of deciduous climbing winding large lianas, as some related research on the chemical composition and pharmacological action of Wisteria sinensis, it’s preliminarily determined that its rhizome leaves contained many kinds of chemical composition such as alkaloids, flavonoids, protein, sugar and glycosides and so on。 Flavonoids has a number of important pharmacological effects, like the activity of inhibition enzyme , anticancer , antiviral, anti-inflammatory, anti-allergic, anti diabetes complications and other functions, it is non-toxic harmless and has effective treatment for some degenerative diseases such as cancer, aging, cardiovascular 。Experimental objective: To research the process of total extraction from Wisteria sinensis by ultrasonic-assisted extraction and investigate the effects on the extraction of the flavonoids in Wisteria sinensis which produced by ultrasonic frequency, ultrasonic time, solid-liquid ratio,ethanol concentration 。 Response surface optimization was used to evaluate the extraction conditions。 Experiment method and content: According to the regression equation of rutin standard curve , the yield of total flavonoids from the Wisteria sinensis is calculated by the absorbance of each dealing with a certain volume of extract and the single factor experiment is carried out to explore the influence of different factors about ultrasonic frequency, ultrasonic time ,solid-liquid ratio and ethanol concentration on the yield of total flavonoids from the Wisteria sinensis。 Then, the response surface method is used to determine the optimal extraction process and evaluate the interaction between different factors。 Experiment results: The optimal experiment conditions is that the ultrasonic time is 46min, the solid-liquid ratio is 1:48 g/mL,the ethanol concentration is 25%。 Under these optimum conditions , the yield of total flavonoids reached as 7。35 mg/g。 Besides, it has scavenging effects on the hydroxyl and superoxide free radicals in a concentration activity。
