摘 要:本文创建了用高效液相色谱法同时测定植物花中的芦丁、槲皮素、儿茶素和橙皮素四种黄酮类化合物的方法。采用乙醇作为提取剂,超声波提取法对迎春花、枇杷花、桂花、野菊花和腊梅花进行黄酮类化合物的提取,并用建立的方法对研究花中的四种黄酮类化合物进行了同时测定。对研究的方法考察显示,用C18柱作为色谱柱,在甲醇-磷酸水溶液(45:55)的流动相下,芦丁、槲皮素、儿茶素和橙皮素分别在0.076~0.38μg(R2=0.9999)、0.064~0.32μg(R2=0.9994)、0.056~0.28μg(R2=0.9996)、0.056~0.28μg(R2=0.9999)内峰面积和进样量呈良好的线性关系,相对标准偏差RSD分别为:1.18%、1.12%、0.79%、1.35%;通过标准加入法得到迎春花提取液中芦丁、槲皮素、儿茶素、橙皮素的平均回收率分别为92.00%(RSD=1.36%)、103.53%(RSD=1.62%)、98.22%(RSD=1.90%)、92.47%(RSD=0.99%)。上述结果标明,本文建立的方法具有较好的精密度与重现性和准确性,可以将四种黄酮类化合物分离。54471

Abstract: This paper has established a method for determination of four kinds of flavonoids, rutin, quercetin, catechins and hesperidin in plant flowers at the same time by high performance liquid chromatography(HPLC). The flavonoids were extracted using ethanol under the ultrasonic condition from the flower of Jasminum nudiflorum, Eriobotrya japonica, Osmanthus fragrans, wild chrysanthemum, Chimonanthus praecox and these were measured by using the method established. The results showed that linear relationship of the peak area and the quantity of rutin, quercetin, catechins and hesperidin were 0.076 ~ 0.38μg(R2 = 0.9999), 0.064 ~ 0.32 μg(R2 = 0.9994), 0.056 ~ 0.28μg(R2 = 0.9996), 0.056 ~ 0.28μg(R2 = 0.9999)respectively by using C18 column and under the mobile phase in the methanol - phosphoric acid aqueous solution (45:55), the relative standard deviation RSD were 1.18%, 1.12%, 0.79%, 1.12% respectively; the average recovery rate of the four kinds of flavonoids were 92.00%(RSD = 1.36%), 1.36%(RSD = 1.62%), 98.22%(RSD = 1.90%), 1.90%(RSD = 0.99%)by added standard four kinds of flavonoids in the extract of Jasminum nudiflorum. The above results indicated that the established method can separate the research of four kinds of flavonoids and has good precision, reproducibility and accuracy.
