摘要:本试验旨在研究日粮添加姜黄素对宫内发育迟缓(intrauterine growth retardation, IUGR)鼠肌肉抗氧化机制的影响。试验采用2×2因子设计,12只正常仔鼠和12只IUGR仔鼠21日龄断奶后随机分成四个处理组(每组6只):NBW组为正常仔鼠,饲喂基础日粮;NC组为正常仔鼠,饲喂基础日粮+400 mg/kg姜黄素;IUGR组为IUGR仔鼠,饲喂基础日粮;IC组为IUGR仔鼠,饲喂基础日粮+400 mg/kg姜黄素。断奶适应期1周,正式试验期4周。结果显示:(1)与NBW组相比,IUGR组大鼠肌肉丙二醛(malondialdehyde, MDA)含量显著升高(P < 0.05),谷胱甘肽过氧化物酶(glutathione peroxidase, GSH-PX)活力显著降低(P < 0.05)。日粮添加姜黄素可显著降低IUGR大鼠肌肉MDA含量(P < 0.05),显著升高总超氧化物歧化酶(totalsuperoxide dismutase, T-SOD)活力(P < 0.05);(2)与NBW组相比,IUGR组大鼠肌肉核因子NF-E2相关因子2(nuclear factor erythroid-2 related factor 2, Nrf2)、超氧化物歧化酶1(superoxide dismutase, SOD1)基因mRNA的表达水平均显著降低(P < 0.05)。日粮添加姜黄素可显著提高IUGR组大鼠肌肉Nrf2、SOD1基因mRNA的表达水平(P < 0.05)。试验结果表明,日粮添加400 mg/kg姜黄素可有效缓解IUGR引起的大鼠肌肉氧化损伤。30508

Effects of curcumin on anti-oxidation in the muscle of IUGR rats
Abstract: The present study was designed to evaluate the effects of dietary supplementation of curcumin on anti-oxidation in the muscle of IUGR (intrauterine growth retardation) rats. Twelve normal and twelve IUGR newborn rats were randomly assigned to the following four experimental groups (n = 6/group): NBW group (normal rats, fed with basic diets), NC group (normal rats, fed with basic diets + 400 mg/kg curcumin), IC group (IUGR rats, fed with basic diets) and IC group (IUGR rats, fed with basic diets + 400 mg/kg curcumin). Rats were fed from 4 to 8 weeks of age,including a one-week adaptive phase for weaning. The results indicated that: (1) compared with NBW group, IUGR rats had a higher content of MDA (malondialdehyde) in the muscle (P < 0.05). Furthermore, the activities of GSH-PX(glutathione peroxidase) was significantly decreased in muscle of the IUGR rats (P < 0.05) as compared with that of NBW group. Curcumin significantly reduced the content of MDA (P < 0.05), whereas increased the activities of T-SOD (totalsuperoxide dismutase) in the muscle of IUGR rats (P < 0.05); (2)compared with NBW group, the mRNA expression of Nrf2 (nuclear factor erythroid-2 related factor 2) and SOD1 in the muscle of IUGR rats were significantly down-regulated (P < 0.05). However, curcumin up-regulated these mRNA expressions (P < 0.05). In conclusion, 400 mg/kg curcumin can effectively alleviate the damage by IUGR and had beneficial effects on the oxidative damage in the muscle of IUGR rats.
