摘要猪流行性腹泻病毒(Porcine Epidemic Diarrhea Virus，PEDV) S 蛋白 COE 区域是参 与机体中和抗体的产生的过程当中十分重要的蛋白区域。将猪流行性腹泻病毒 (Porcine Epidemic Diarrhea Virus，PEDV) S 蛋白 COE 区域在原核表达系统 DE3 当中 进行表达，并对获得的蛋白质进行 SDS-PAGE 鉴定，从而在体外获得一定量的 COE 蛋白。83569
根据 coe 基因进行正向引物和反向引物的设计，添加 BamHⅠ单酶切位点并添加 一段与载体 pET28(a)质粒同源的序列，利用 PCR 反应扩增 coe 基因，线性化 pET28(a) 质粒载体，通过同源重组连接将 coe 基因连接到载体上。BamHⅠ单酶切鉴定重组质 粒，用单酶切鉴定正确的 PEDV coe-pET28(a)重组质粒转化 TOP10 感受态细胞，挑取 单菌落培养并进行菌液 PCR，对阳性克隆进行重组质粒的抽提，将得到的质粒寄送测 序公司测序。将测序鉴定正确的 PEDV coe-pET28(a)重组载体转化到表达菌 BL21 中， 用 IPTG 诱导表达，终浓度为 1。0 mmol/L，分别在 2h、4h、6h 时收集菌液。制备样品 进行 SDS-PAGE 分析，最终在约 20KDa 处发现一条特异表达条带，该结果与理论预 期相符合，为进一步研究 PEDV coe 基因的功能以及制备 PEDV 相关疫苗奠定一定的 基础。

Abstract The coe regional of Porcine Epidemic Diarrhea Virus(PEDV) spike（S）takes a great roll on the inducement of neutralizing antibody in animals。 Express the coe gene of PEDV in the DE3 cell, identify the protein by SDS-PAGE and get some COE protein in vitro。
Design primers, add BamHⅠRestriction Enzyme Cutting site and homologous
sequences of pET28(a) to the primers, linearizing the pET28(a) plasmid, joining coe gene with pET28a(+) vector by homologous recombination。 Identify the recombinant plasmid with BamHⅠ, transfer the right recombinant plasmid into competent top10 cell, identify the recombinant plasmid by bacteria liquid PCR, extracting recombinant plasmid and sequencing it。 Transfer the recombinant plasmid with right sequence into BL21 and express it with 1。0 mmol/L IPTG。 Gathering the bacteria liquid after 2h,4h and 6h, identify the
protein by SDS-PAGE, a 20KDa expression stripe is found, which is in line with the expectations。
