摘要:稻米垩白率和直链淀粉含量是影响稻米食品质的两个重要因素,垩白率受到多基因控制以及环境因素的影响,直链淀粉的含量主要和Wx有关系。染色体片段置换系基因组内只含有一个或几个来自供体亲本的染色体片段,可用于QTL鉴定、精细定位、图位克隆和品种改良利用,是分子育种的良好材料。本实验利用9311/普通野生稻的染色体片段置换系,通过检测稻米垩白率和直链淀粉含量,发掘群体内高垩白以及高/低直链淀粉含量的家系,并结合分子数据寻找控制两种性状的QTLs。实验发现,(1)2016年收获的材料垩白率普遍偏低,经分析认为可能是在稻米灌浆时期持续降水导致光合产物偏低,稻米灌浆变慢,使得胚乳中的淀粉体和蛋白体缓慢发育,垩白发生率降低;(2)通过测定稻米的直链淀粉含量,筛选出2份高直链淀粉含量的材料和1份低直链淀粉含量的材料,分别为Q187、Q110和Q87,直链淀粉含量分别为20.3%、21.1%和2.9%,其中受体亲本9311的直链淀粉含量为14.5%;(3)经Western blot验证,发现Q87为Wx基因缺失突变,导致米质呈糯性,直链淀粉含量降低,Q187和Q110为Wx基因蛋白水品上调,直链淀粉含量升高。31405

Detection of QTL for chalkiness and amylose content by using chromosome segment substitution lines of 9311 /O.Rufipogon in rice
Abstract:Chalkiness and amylose content are two important factors in rice which affecting the cooking and eating quality of rice, they are effected by environment and multiple genes.Chromosomal segment substitution lines(CSSLs),in which each line carries a single or a few chromosomal segments from a donor in the genetic background of recurrent are perfect materials for QTL identification,fine mapping and map-based cloning of QTL,variety improvement and soon.By detecting rice chalkiness and amylose content , find the mutants with high chalkiness and high / low amylose content in 9311 /O.Rufipogon CSSLs,to looking for the QTLs controling those two characters.The results showed that (1)the chalkiness rate of material harvested in 2016 was lower then last year.Persistent precipitation during the grain filling stage may reduced photosynthetic products of rice,result in the grouting getting slower ,so the amyloids and proteosomes in endosperm growth slowly,which cause the chalkness getting lower.(2)We selected 2 high amylose content materials and 1 low amylose content material during the measuring, they are Q187, Q110 and Q87 which amylose content was 20.3%, 21.1% and 2.9%, and the parant of 9311 was 14.5%.(3)Spontaneous mutation of Wx of Q87 caused it become glutinous rice, so the amylose content decreased. Protein level 偶of Wx gene enhancement in Q187 and Q110 resulted amylose content increased.

利用9311/普通野生稻的染色体片段置换系定位控制水稻垩白率和直链淀粉含量的QTL
引言:水稻是世界上最主要的粮食作物之一。长期以来,随着人口增长、环境问题以及国民粮食安全问题,在水稻育种和生产工作中,提高水稻产量一直是重中之重。但是近年来,随着国民生活水平的提高,人们对稻米品质要求也在不断提高。此外,国际粮食市场的竞争十分激烈,也促使了我国水稻育种目标从片面的高产转向高产与优质并举。因此稻米品质的遗传学研究成为水稻研究中重要的一部分。实验利用以9311为背景的9311/普通野生稻的染色体片段置换系214个家系作为实验材料,将鉴定其垩白率和直链淀粉含量的两项重要稻米品质性状表型值,通过调取CSSL群体材料的分子连锁图谱,寻找控制两种性状的基因片段所在位置。 用9311×普通野生稻的染色体片段置换系定位控制水稻垩白率和直链淀粉含量的:http://www.youerw.com/shengwu/lunwen_27554.html
