摘 要:木霉的ACCD酶可以切断植物激素乙烯的前体ACC产生酮戊二酸和氨,降低植物的乙烯含量从而促进植物生长,提高植物的抗病性。采用硫酸氨分级沉淀的方法对来自木霉ACCD酶进行了分离纯化,然后用不同浓度的ACCD酶处理大豆植株。结果表明ACCD酶对大豆具有明显的促进作用。大豆的生长指标(根长,株高,茎粗,干重和湿重)和生理指标(POD活性,SOD活性,CAT活性)在ACC浓度为0.0005g/ml,0.001g/ml,0.0015g/ml和0.002g/ml时依次缓慢上升,在0.002g/ml的浓度处达到最高值,在0.002g/ml浓度之后的0.0025g/ml和0.003g/ml大豆的生长指标和生理指标表现为逐渐下降。关键词:木霉;ACCD;分离;纯化;功能8260
Separation, Purification and Function of Amino Cyclopropane- 1-1-carboxylic Acid Deaminase(ACCD)from Trichoderma
Abstract: Amino cyclopropane-1-1-carboxylic acid deaminase (ACCD) fromTrichoderma could cut the ethylene precursor (ACC) of plant hormone to produce ketoglutaric acid and ammonia to, and then lower ethylene content and promote plant growth, furthermore, to improve the disease resistance of plants. At present, ACCD enzyme was separated and purified using the ammonium sulfate fractionation method. Then ACCD was used to treate soybean plants. It was showed that the ACCD enzyme had a significant effect on soybean growth. The growth indexes , such as root length, plant height, stem diameter, dry weightand wet weight, as well as resistant enzymes indexes (POD activity,SOD activity, CAT activity) rised slowly with the ACC concentration rising, and the best optimal concentration was 0.002g/ml. When the concentration reached0.0025 g/ml the rising tends of soybean growth index and physiological index declined gradually.
