摘要从江苏连云港沿海滩涂采集的盐生植物中分离得到内生放线菌KLBMP 5180,前期研究发现该菌株具有对宿主植物较好的解盐促生作用,盐胁迫下能够显著促进植物的生长。本论文采用细菌多相分类学方法对其进行详细的分类鉴定,通过形态学观察、生理生化、化学分类以及16S rRNA基因序列系统发育分析等方面的研究,确定菌株KLBMP 5180是好氧性革兰氏阳性菌。菌株的最适生长温度范围为4-37oC,最适pH范围为pH4-10,最适NaCl浓度为0-7% ,可以在多数培养基上良好的生长,菌落呈浅黄色,无菌丝形成。主要的极性脂类型为DPG、 PG、PI、PL。主要的甲基萘醌类型为MK-9,MK-8和MK-8(H2)。菌株KLBMP 5180的(G+C)mol%为60 %。基于16S rRNA基因序列分析显示菌株KLBMP 5180与A. arilaitensis Re117T 的亲缘关系最近,基因序列相似性为99.26%。以上结果表明菌株KLBMP 5180应是放线菌门中节杆菌属中的一个新物种,拟命名为盐生植物节杆菌(Arthrobacter halophytocola sp. nov )。71133

Study on the Taxonomy of a Salt Tolerant and Plant Growth Promoting Arthrobacter Strain
Abstract The endophytic actinobacterial strain KLBMP 5180, which was isolated from the inner tissues of a halophyte collected from Lianyungang coastal area, was found to have plant growth promoting activity and promoted the host plant growth significantly under salt stress. In this paper, the detailed taxonomic status of this strain was studied by using the bacterial polyphasic taxonomy methods, including morphological, physiological and chemotaxonomic characteristics, and the 16S rRNA phylogenetic analysis. The optimum growth temperature range is 4-37oC, the optimum pH range is pH 4-10 and the optimum NaCl concentration for growth is 0-7%. Strain grew well on most tested media with light yellow colonies and no hyphae. The main polar lipids were DPG, PG, PI and PL. The major menaquinones were MK-9, MK-8 and MK-8 (H2). The G+C mol% content is 60 %. Based on 16S rRNA gene sequence analysis, strain KLBMP 5180 showed highest gene sequence similarity of 99.26% with type species A. arilaitensis Re117T. The combined results showed that strain KLBMP 5180 should be assigned as a new species of the genus Arthrobacter, for which the name Arthrobacter halophytocola sp. nov. is proposed.
