摘要在本工作中,我们将有机显色剂与金属离子形成的螯合物用作比色探针,实现了对碱性磷酸酶(Alkaline phosphatase, ALP)活性的简便、快速、高灵敏和高选择性比色分析。由于ALP能将底物抗坏血酸-2-磷酸水解产生抗坏血酸,生成的抗坏血酸可以将Fe3+还原成Fe2+,后者与水溶性红菲绕啉配体结合形成的络合物呈红褐色并且在波长535 nm处有很强的光吸收,因此可用于ALP活性的定量比色分析。基于溶液颜色变化或紫外-可见吸收光谱,我们实现对0-220 mU mL-1范围内ALP活性的快速、高灵敏比色分析。同时,该方法用于检测ALP活性具有很高的选择性,并可用于筛选相关ALP抑制剂。此外,该方法也可直接用于血清或全血样品中ALP活性的快速、高灵敏比色分析,因此具有很好的实用性和临床应用前景。78509

Title Colorimetric assay of alkaline phosphatase activity and inhibitor screening
Abstract In this work,a simple and fast method has been proposed for the sensitive and selective colorimetric assay of alkaline phosphatase (ALP) by exploiting a complex of organic chromogenic agent and metal ion as the probe。 In the presence of ALP, ascorbic acid 2-phosphate is dephosphorylated to produce ascorbic acid, which in turn reduce Fe3+ to Fe2+。 As the complex formed between Fe2+ and bathophenanthroline is red-brown in color and featured by an intense absorption band at the wavelength of 535 nm in the visible region, it can be taken advantage of for the colorimetric assay of ALP activity。 Based on the change in solution color or UV-Vis absorption spectrum, the ALP activity in the range from 0 to 220 mU mL-1 has been sensitively and rapidly detected。 The results indicated that this method is highly selective, and also applicable for the screening of ALP inhibitor。 In addition, it shows a good analytical performance when applied for the colorimetric assay of ALP activity directly in complicated serum or whole blood samples。 Therefore, it holds great potential in clinical applications。
