葛根素抑制A549细胞迁移研究

目的:以肺腺癌A549细胞为模型,检测葛根素对A549细胞增殖、迁移和存活等方面的影响,以此研究其作用机制,进一步扩展葛根素在肺癌领域的研究,并为其应用到实际方面提供实验依据。

方法:通过划痕试验,观察细胞在不同浓度下的迁移情况;检测培养液上清清除自由基的水平;流式细胞仪检测细胞周期76478

内容:掌握A549细胞的培养和传代;明确葛根素对肺腺癌A549细胞的影响;检测葛根素抗氧化能力及对A549细胞周期的影响。

结果:在倒置显微镜下观察到,葛根素可以抑制A549细胞的生长,且随葛根素浓度增高,抑制更加明显;划痕实验中,相对于溶剂对照,葛根素显著抑制A549细胞的迁移,且随P浓度升高,细胞迁移率越小;自由基清除实验结果显示,葛根素并未改变A549细胞中的氧化状态;进一步分析细胞周期分布的变化,发现葛根素显著升高了S期的细胞比例,表明其可能将A549细胞阻滞在S期,进而抑制细胞的生长。

结论:葛根素靶向A549细胞,将其细胞周期阻滞在S期,抑制细胞的生长和迁移;这一过程并不涉及葛根素的抗氧化活性。

Research of puerarin on inhibition of A549 cell migration

Abstract Objective:This paper uses the lung cancer A549 cells as a model to detect the effects of puerarin on proliferation, migration and survival and so on and aims to elucidate the possible mechanisms, to further expand the research of puerarin in the field of lung cancer and provide experiments evidence for their application。

Methods: Observation of cell migration after puerarin treatment by scratch test; Detection of anti-oxidation activity in culture supernatant; Analysis of cell cycle by flow cytometry。

Content: Master the culture and passage of A549 cells; Confirm the effects of puerarin on lung cancer A549 cells; Detect the elimination of free radicals; Test the cell cycle distribution。

Results: The proliferation of A549 cells was inhibited by puerarin as observed under the inverted microscope, and the inhibitory effect was more obvious in puerarin-treated group with high concentration。 In the scratch test, compared with DMSO group, the cell migration was suppressed significantly by puerarin in dose-dependent manner。 However, no alteration of anti-oxidation activity was detected in both control and puerarin group。 The result of flow cytometry showed that the ratio of cells in S phase was increased by puerarin in comparison with control group。

Conclusion: Puerarin inhibits the migration of A549 cells and blocks A549 proliferation into S phase。 The anti-oxidative property of puerarin is not involved in this process。
