摘要:华鳈(Sarcocheilichthys sinensis)隶属于鲤科(Cyprinidae)、鮈亚科(Gobioninae)、 鳈属(Sarcocheilichthys),是一种生活于水体中下层的小型鱼类。Hepcidin(heppatic bactericidal protein)是鱼类的一种重要抗菌肽,它具有杀菌活力高、作用迅速、不产生耐 药性等特点,是多细胞生物免疫防御的有效手段。本研究采用 RT-PCR 方法,从华鳈(Sarcocheilichthys sinensis)中克隆获得了 Hepcidin cDNA 序列,并对其进行了同源性比 较和进化分析。结果显示,华鳈 Hepcidin cDNA 片段长为 654 bp,开放阅读框长 276 bp, 编码 91 个氨基酸。与近源物种的氨基酸相似性较高,并存在较多氨基酸保守区域。本研 究获得的华鳈 Hepcidin 基因序列将为今后在华鳈中开展该基因的多态性位点与抗病性状的 关联分析、免疫机制与免疫性能研究、抗病品种的分子标记辅助选育等提供基础资料。85859

Abstract : Chinese fat minnow ( Sarcocheilichthys sinensis ) belonging to the genus Sarcocheilichthys in subfamily Gobioninae of the Cyprinidae family is a tiny fish lives at the lower layer in water。 Hepcidin is an important antibacterial peptide in fish。 It has characteristics of high bactericidal activity, rapid action, no drug resistance, and so on, therefore, it plays an significant role in immune defense system of multicellular organisms。 In this present study,
Hepcidin gene was amplified from the Chinese fat minnow (Sarcocheilichthys sinensis) by
RT-PCR, meanwhile, homologous and phylogenetic analyses were also performed。 Finally, a cDNA fragment of Hepcidin with a total length of 654 bp was obtained, and the open reading frame 276 bp, which encoded a peptide containing 91 amino acids。 Compared to close-related fishes, the similarity rate of S。 sinensis Hepcidin was quite high with large regions of conserved amino acid sequences。 The cDNA fragment of Hepcidin obtained in this study will be useful tools for further researches on association analysis between polymorphic sites and disease-resistance abilities, immune mechanism analyses, marker assisted selection breeding for new disease-resistance broodstocks in this fish。
