摘要:原代培养是指在体外仿造体内的生理环境,让从体内取出的组织细胞生存。生长和传代,并维持原有组织细胞功能和结构特性。目的:本课题将对鸽嗉囊成纤维细胞体外培养建立方法,目的在于为今后研究鸽嗉囊成纤维细胞与上皮细胞之间的相互作用以及嗉囊上皮细胞形成鸽乳物质的机理打下基础。方法 :利用胰酶细胞消化法和组织块贴壁法培养鸽嗉囊成纤维细胞,并在光学显微镜下观察细胞形态,用MTT法检测细胞活力,用免疫荧光法鉴定细胞类型。结果:胰酶消化法和组织快贴壁法培养下的嗉囊组织成纤维细胞的形态正常,符合一般成纤维细胞的梭形或纺锤形结构特征,细胞的增殖与活性良好,利用Vimentin抗体经免疫荧光检测为阳性,说明上述培养细胞为典型的成纤维细胞。结论:本研究利用胰酶消化法和组织块贴壁法成功建立了成纤维细胞的培养模型。94541

Abstract: primary culture is to mimic the physiological environment in vivo, so that the tissue cells removed from the body survive, grow and reproduce, and maintain the functional and structural characteristics of the original tissue cells。 Objective: this topic to the pigeon crop cultivation method of fiber cells in vitro, to foster mechanism model of pigeon milk lays the foundation for the establishment of material pigeon fibroblast crop tissue adherent cells digestion method for the future research of pigeon crop interaction between fibroblasts and epithelial cells and epithelial cells。 Methods: cells by trypsin digestion and tissue explant culture method of pigeon crop fibroblast cells, and the cell morphology was observed under optical microscope, cell viability was measured by MTT method, by the method of immunofluorescence to identify the cell types。 Results: by the method of trypsin digestion and tissue adherent culture, crop tissue fibroblasts showed the normal spindle shape from fibroblast cells。 Cell proliferation and activity were also detected by MTT method。 By immunofluorescence of vimentin antibody, the result was positive that the cultured cells were typical fibroblast。 Conclusion: the culture model of fibroblasts was successfully established by trypsin digestion and tissue block adherence method。
