摘要目的:研究木犀草苷(luteoloside)对肝癌Huh-7细胞迁移和侵袭能力的影响,并分析其相关机制。方法:划痕实验观察木犀草苷对Huh-7细胞转移的影响;Transwell实验观察木犀草苷对Huh-7细胞迁移和侵袭能力的影响;ROS水平检测考察木犀草苷对Huh-7细胞内活性氧水平的影响;采用Western blot方法检测Huh-7细胞中NLRP3、caspase-1和IL-1β的表达变化情况。结果:在划痕实验中,未经木犀草苷处理过的空白对照组的损伤宽度区域明显小于处理组;Transwell实验结果表明,经木犀草苷处理的Huh-7细胞的穿膜数量明显少于空白对照组,迁移实验和侵袭实验都有相似的效果;蛋白水平检测发现Huh-7细胞内NLRP3、Caspase-1和IL-1β蛋白表达水平明显下降。在本研究中,我们购买的是从药用植物中提取出来的木犀草苷。一系列实验表明,木犀草苷对肝癌细胞的转移具有明显的抑制作用,这将为治疗肝癌疾病提供新思路。69850

Luteoloside Suppresses Metastasis of the Liver Cancer Cells by Inhibition of Inflammasome
Abstract
Objective: To study the effect of luteoloside on cell migration and invasion of Huh-7 cells, and to analyze its mechanism. Methods: We observe the effect of luteolin on Huh-7 cell metastasis by the wound healing experiment; We observe the influence of luteololoside on invasion and migration of Huh-7 cells by transwell experiment; We observe the influence of luteololoside on reactive oxygen species level in Huh-7 cell by detection of ROS; Using Western blot method detect the expression of NLRP3,caspase-1 cells and IL-1β. Results: In the wound healing experiment , The width of the damage zone was larger than that of the blank control group; In the transwell experiment, the number of transmembrane cell were significantly less than the control group, the migration experiment and invasion test have similar effects. Through the protein detection, it was found that the level of NLRP3、Caspase-1 and IL-1β expression in Huh-7 cells was obviously decreased. In this study, we bought luteoloside which is extracted from medicinal plants. A series of experiments show that luteoloside has obvious inhibitory effect on liver cancer cell metastasis, which will provide new idea for the treatment of liver diseases..
