摘要:本论文以绞股蓝(Gynostemma pentaphyllum)为原料,提取得到了绞股蓝多糖(Gynostemma pentaphyllum polysaccharides,GPP),优化了提取条件,并进行了纯化。采用热碱水浸提的方法从绞股蓝中提取活性多糖,通过正交设计试验确定了热水浸提绞股蓝多糖的最佳浸提工艺条件:料液比为1:20;加热温度为80℃;加热时间为3h。采用Sevag法脱蛋白,这种方法采用氯仿—正丁醇溶液除蛋白质,优点是条件温和,随着脱除次数的增加,蛋白质含量逐渐降低。通过DEAE-32离子交换柱层析进一步分离,然后再用Sephacryl S-400分子筛进行纯化,最后测定了绞股蓝多糖的分子量和纯度。6664

Extraction and Purification of the Gynostemma Pentaphyllum Polysaccharides
Abstract: Gynostemma pentaphyllum as raw material, this paper got Gynostemma pentaphyllum polysaccharides extraction, extraction condition was optimized, and purification was carried out. Adopt the method of hot alkaline leaching extract activity of polysaccharide from Gynostemma pentaphyllum. By the orthogonal design experiment of Gynostemma pentaphyllum polysaccharides, hot alkaline water leaching extraction was determined as the best extraction technology conditions: material liquid ratio of 1:20; The heating temperature is 80 ℃; The heating time for 3 h. Off protein by Sevag method, this method uses the chloroform and N-butanol solution in addition to protein, advantages of mild conditions, with the increase of removal times, protein content gradually reduced. By DEAE Cellulose DE - 32 further separation, ion exchange column chromatography and then use Sephacryl S - 400 molecular sieve purification, the determination of the molecular weight and purity of gynostemma polysaccharide.
