摘 要:具有手性药物的重要性在新药中越来越受到重视。某种具有特定功能基团的手性醇[2]也是合成手性药物和其它手性化学品的重要中间体。手性醇[2]合成方法的成熟与改进具有积极的促进手性酶的作用。酵母细胞催化芳基酮的不对称还原反应正交试验综合考察了细胞量 、添加剂、底物浓度和底物浓度的量等因素对反应的影响。同时,考察了芳香酮[3]结构对产物对映选择性的影响 , 并且还进行了物料衡算和热量衡算来设计,为了达到更好转化率。同时在实验中发现对映选择性的变化规律符合Prelog规则[4],与羰基相连的两个基团体积差异越大,对映选择性越好,这类反应已被广泛应用于医药、化工、食品等行业。目前,生物法还原芳基酮[2]化合物处于研究领域不断扩大和工业应用迅速发展的阶段。9296

Studies on reduction of aryl ketones catalyzed by yeast cells
Abstract: The importance of chiral drugs in new drugs[2] have been paid more and more attention. Some important intermediates of chiral alcohols with specific functional groups and chiral synthesis of chiral drugs and other chemicals[2]. Play a positive role in promoting and improving method of chiral enzymatic synthesis of chiral alcohols maturity. Orthogonal test of yeast cell asymmetry catalytic reaction of aryl ketone reduction in analyzing the effects of cell volume, additives, substrate concentration and substrate concentration on the reaction rate[4] .At the same time, the aromatic ketone structure influence on the enantioselectivity of the product, and the material balance and heat balance calculation of design, in order to achieve better conversion rate. Also found in the experiments with Prelog rules change law of the enantioselectivity of two groups, the difference in volume and carbonyl is bigger, enantioselectivity is better, this kind of reaction has been widely used in pharmaceutical, chemical, food industry etc.. At present, the biological reduction of aryl ketone compounds in the research field of expanding and industrial application of rapid development stage.
