小分子药物Adjudin调控肺鳞癌细胞自噬的研究

摘要djudin，或称 AF-2364，是一种广谱抗癌药物 lonidamine(LND，氯尼达明)的同系物。Adjudin 最初是开发作为潜在的非荷尔蒙男性避孕药，但它的其他药理活性也不断被发现。之前有研究表明，Adjudin 对多种癌细胞具有杀伤作用，同时与顺铂（临床上常用的化疗药物）联用，可以明显提高多种癌细胞对顺铂的敏感性。我们立足于肺鳞癌，用 CCK-8 法测定 Adjudin 对肺鳞癌细胞的杀伤作用，同时使用 Western blot 和瞬转 GFP-LC3 质粒两种方法探测 Adjudin 对肺鳞癌细胞自噬水平的影响。结果表明， Adjudin 对肺鳞癌细胞具有一定的杀伤作用，并能影响肺鳞癌的自噬水平。73301

Title The effect of Adjudin on autophagy of squamous cell lung carcinoma

Abstract Adjudin,also known as AF-2364, is analog of broad-spectrum anti-cancer drugs lonidamine(LND)。 Adjudin was originally developed as a potential non-hormonal male contraceptive, but its other pharmacological activities were also found gradually。 Prior studies have shown that Adjudin has killing effect on a variety of cancer cells。 In conjunction with cisplatin (chemotherapy drugs commonly used clinicallly), Adjudin can significantly improve the sensitivity of a variety of cancer cells to cisplatin。 Based on lung squamous carcinoma, the effects of Adjudin on lung squamous cell viability were assayed by CCK-8 method, and autophagy levels were determined by Western blot and fluorescent protein labeling。 The results show that Adjudin could kill lung squamous carcinoma cells, and influence the autophagy level of lung squamous carcinoma cells。
