摘要重阳木属于暖温带速生大戟科树种。大戟科植物含有黄酮,具有抵抗肿瘤、抵抗病毒、抵抗氧化、抵抗炎症等多种生物学活性。在本次试验中,我采用超声波萃取法对重阳木叶的黄酮乙醇提取工艺进行优化。采用响应曲面法将连续变量曲面模型建立起来,对影响重阳木黄酮萃取过程的因子及其交互作用来进行评价,确定最佳水平范围。结果表明:重阳木叶中总黄酮的最佳超声提取工艺为体积分数39。5%乙醇溶液、液料比1:47。3(g/ml)、超声功率70 W、萃取时间51。5 min。最后用LC-MS法检测重阳木提取黄酮的主要黄酮类型及结构特点,测定其体外抗氧化能力。通过对重阳木叶中总黄酮的提取液进行体外抗氧化性实验,表明其具有较强的抗氧化活性,尤其对清除羟自由基有着较强的作用,且其抗氧化性随着黄酮含量的增加而有着明显的增加,具有进一步研究利用的价值。75034

Optimization of Ultrasonic Assisted Extraction of Total Flavonoids from the Leaves of Bischofia polycarpa by Using Response Surface Method
Abstract Bischofia polycarpa grows in the warm temperate zone。 It belongs to Euphorbiaceae fast-growing tree species。 Euphorbiaceae plant contains flavonoids。 It has a variety of biological activity of antioxidant, including anti-tumor, anti-viral, anti-oxidation, anti-inflammatory。 This test uses ultrasonic extraction of flavonoids from Bischofia polycarpa to process the optimization of ethanol extraction and use response surface methodology to establish a continuous variable surface model to evaluate factors which affects biological processes and their interactions to determine the optimum level range。 The optimum conditions for extracting total flavonoids from the leaves of Bischofia polycarpa were found that ethanol concentration 39。5 %, material-to-liquid ratio 1:50 (g/mL), ultrasonic power 70 W and length of ultrasonic treatment of 51。5 min。 Finally, detecting major types and structures of flavonoid by LC-MS, measured its capacity of vitro antioxidant。 The total flavonoids from the leaves of Bischofia polycarpa had obvious anti-oxidation property, especially in scavenging effects on hydroxyl free radicals。 And the antioxidant ability increased in a concentration-dependent fashion。
