猪卵母细胞IVM和精子质量对胚胎发育影响的研究

摘要:本实验采用卵母细胞体外成熟、精子获能、体外受精及胚胎培养技术,通过对照重复试验来探讨猪精子不同的获能方法、精子活率及卵母细胞的选择对猪体外受精后卵裂率和胚胎发育的影响。结果表明,同一猪种精液采用不同的获能液,体外受精后卵裂率无显著差异(P>0.05),但各组的桑葚胚率,三元猪+获能液A组明显高于三元猪+获能液B组(P<0.05),而新淮猪的两组情况相反;精子活率的三组实验的卵裂率和桑葚胚率差异显著(P<0.05),且随精子活率的降低而降低;体外成熟前卵母细胞外的卵丘细胞层数<3层,体外受精后的卵裂率和桑葚胚率都明显低于卵丘细胞层数≥3层的卵母细胞(P<0.05)。25685 毕业论文关键词:卵母细胞;精子获能;体外受精 The study of the effects of oocytes maturation and sperm quality on the developmental viability of pig embryos Abstract:In the study,techniques including in vitro maturation of oocytes,capacitation of sperm ,in vitro fertilization and embryo culture were used to research the effects of different methods of capacitation of sperm, sperm viability and selection of oocytes on cleavage rate and embryo development .Results show that no significant difference in the cleavage rate of the same pig semen with different emollients(P>0.05),the morula rate was significantly higher in the Sanyuan pig+Capacitation fluid A than in the Sanyuan pig+Capacitation fluid B(P<0.05).Subsequently, semina from Xinhuai with different sperm viability were performed for the IVF. And results show that significant differences in cleavage rate and morula rate were observed in fertilized embryos between different sperm viability(P<0.05).Finally,the cleavage rate and morula rate in the COCs with less than 3 layers were significantly lower than in the COCs with more than 3 layers(P<0.05).
