西红花组织培养技术研究

摘要:目的:确定西红花球茎、幼叶愈伤组织诱导的适宜激素配比,为西红花的组织培养、快繁等研究奠定基础和科学依据。方法:以西红花球茎部位为材料,以MS为基本培养基,以激动素(KT)、吲哚丁酸(IBA)、萘乙酸(NAA)作为单因素变量,确定诱导西红花球茎愈伤组织诱导的适宜条件;以MS+0.5 mg•L-16-BA为基本培养基,添加不同浓度的2,4-D作为变量,确定诱导西红花幼叶愈伤组织形成的条件。结果:在仅含有KT的培养基中,诱导球茎形成愈伤组织效果最好;且浓度为0.2 mg•L-1KT的诱导率最高,诱导率为71.00%。在激素组合中,诱导西红花幼叶产生愈伤组织的最佳激素组合是MS+0.5 mg•L-16-BA+1.2 mg•L-1 2,4-D。诱导率为67.00%。结论:在单种植物生长调节剂的处理下,KT诱导西红花球茎形成愈伤组织的诱导率高。在幼叶诱导形成愈伤组织中,以MS+0.5 mg•L-16-BA+1.2 mg•L-1 2,4-D 培养基效果最好,诱导率最高。30502

Study on the tissue culture of Crocus sativus
 [Abstract] Objective: To study the most suitable hormone and the concentration of callus induced with the young leafs and corms of Crocus sativus, laying the foundation and scientific basis for the breeding of saffron cultivation. Method: The corms of Crocus sativus cultured in the medium that was MS supplemented with single plant growth regulator, as kinetin (KT), indole butyric acid (IBA) and 1-Naphthylacetic acid (NAA), to determine the appropriate conditions of hormone induced callus of saffron. The young leafs of Crocus sativus cultured in the medium which was added 0.5 mg•L-16-BA of MS supplemented with different concentration of 2,4-D, to determine the appropriate conditions of hormone induced callus of saffron. Result: In the study of induced callus with corms of Crocus sativus, it was the best medium of KT, and the induction rate was 71.00% at the concentration of 0.2 mg•L-1. In the study of hormone combination , saffron Leaves Callus Induced by hormone combination was the best MS+0.5 mg•L-16-BA+1.2 mg•L-1 2,4-D. The induction rate was 67.00%. Conclusion: In the treatment of single plant growth regulator induced callus, the medium with KT induced the highest induction rate. In young leafs callus induction, the medium with MS+0.5 mg•L-16-BA+1.2 mg•L-12,4-D had the best effect and the highest induction rate.
