“蓝香水”月季组织培养技术的研究

摘要:以“蓝香水”月季带腋芽的茎段为研究对象,研究不同的处理对月季快繁、愈伤组织诱导、增殖、再生和生根的影响。结果表明:在MS+6-BA 1.5mg/L+NAA 0.05mg/L培养基月季芽增殖倍数达到5.0;月季试管苗在加入活性炭3.5g/L+抗坏血酸 0.5mg/L的培养基上发芽率、成活率最高,分别为94.4%、93.5%;外植体在MS+6-BA 2.0mg/L+NAA 2.0mg/L+2,4-D 1.0mg/L上,茎段平均诱导率最高达93.1%;愈组织在MS+6-BA 0.5mg/L+NAA 0.5mg/L+2,4-D 2.0mg/L培养基上诱导率最高达到90.4%,且长势最好;愈伤组织继代增殖在MS+6-BA 1.0mg/L+NAA 0.05mg/L培养基上增殖率达97.7%;最适不定芽诱导培养基为MS+6-BA 1.5mg/L+NAA 0.1mg/L+2,4-D 2.0mg/L,诱导率可达86.7%最高;最佳生根培养基为1/2MS+6-BA0.10mg/L+NAA0.2 mg/L+活性炭0.5g/L,生根率达91.6%。39165 毕业论文关键词:月季;组织培养;愈伤组织;再生 Study on Plant Tissue Culture Technology of Chinese Rose Abstract:Taking "Blue perfume"Rose with axillary buds as the experimental material ,different treatments for rose propagation,callus induction,proliferation,regeneration and rooting were explored.The results showed that:on the medium MS+6-BA1.5mg/L +NAA 0.05mg/L,the rose bud’s multiple of regeneration was 5.0;Rose plant lets on activated carbon was added 3.5g/L+Vc 0.5mg medium/L of germination rate,survival rate up to,respectively,94.4percent、93.5percent;explants on MS+6-BA 2.0mg/L+NAA 2.0mg/L+2,4-D 1.0mg/L, the average induced stem section was the highest rate of 93.1 percent;callus in MS+6-BA 0.5mg/L+NAA 0.05mg/L+2,4-D2.0mg/L medium induced rate was90.4percent,and the best growth;callus subculture in MS+6-BA1.0mg/L+NAA A 0.05mg/L medium on proliferation rate was 97.7%;the optimal medium for adventiti -ous buds MS+6-BA 1.5mg/L+NAA0.1mg/L+2,4-D2.0mg/L,the highest induction rate was 86.7 percent;the best medium for rooting was 1/2 MS+6-BA 0.10mg/L+NAA 0.2 mg/L+activated carbon0.5g/L,the rooting rate was 91.6%. Keywords:Rose;tissue culture;Callus;regeneration 目录 摘 要1 引言 2 1.材料与方法3 1.1实验材料3 1.2实验方法...3 2.结果与分析4 2.1不同处理对月季芽增殖的影响4 2.2不同抗褐变剂对月季试管苗生长状况的影响5 2.3不同外植体部位选取对月季愈伤组织的诱导的影响...6 2.4不同处理对月季愈伤组织诱导的影响.6 2.5不同处理对月季愈伤组织继代增殖的影响7 2.6不同处理对月季不定芽诱导的影响7 2.7不同处理对月季生根的影响7 3.结论与讨论8 参考文献9 致谢...11 月季组织培养技术的研究引言 月季(Rosa chinensis)别名斗雪红、长春花、月月红、瘦客等,是蔷薇科(Rosaceae)蔷薇属(Rosa L.)常绿或多年生落叶灌木植物,是我国十大传统名花之一,同时也是世界上著名的四大切花之一[1]。月季原产于我国,17世纪传入欧洲,现已遍布世界各地。由于其特殊的情感内涵和商品价值,被广泛应用于园林、庭院装饰,并可制成月季盆景、切花、花束、花篮等。另外,月季花可提取香料,叶、根、花均可入药,具有活血消炎、消肿解毒等功效[2]。Hasegawa[3]于1980年成功的在MS培养基上建立了月季试管苗无性系,开辟了月季组培的先河。 有很多研究人员在探讨影响月季离体快繁因素时发现,影响因素是多方面的,其中主要影响因素有激素的种类及浓度、抗褐变剂种类及浓度、外植体选择等。陈建雄[1]以月季腋芽为外植体进行离体快繁,在MS+6-BA 2.0mg/L+NAA 0.5~1.0 mg/L+GA 1.0mg/L培养基上,分化率为95%,并将分化的幼芽转移到1/2MS+NAA 0.5~1.0mg/L培养基上诱导生根,成活率达95%以上。宋丽莎[4]等以盆栽微型月季“粉和平”为试验材料,得出适于诱导芽分化的培养基为MS+6-BA 2.0mg/L+NAA 0.1mg/L,适于增殖的培养基为MS+6-BA 3.0mg/L+NAA 0.3mg/L。卢绪娟[5]以“中国红”月季枝条为试验材料,抗褐变剂以活性炭3.0g/L对月季水培扦插成活状况的处理效果最佳。冯欢[6]等选取微型月季幼嫩离体叶片为外植体,愈伤组织在培养基MS+6-BA 1.0mg/L+NAA 0.1mg/L+TDZ 0.02mg/L上的再分化率达到92.6%。项艳[7]等以“红成功”带侧芽的茎段为实验材料,得出在MS+6-BA 2.0mg/L+NAA 0.05mg/L+活性炭0.3%+蔗糖30g/L培养基上对茎段分化和植株再生效果最好,最适于长根的培养基是MS+NAA 0.05mg/L+活性炭0.3%+蔗糖30g/L。周晓馥[8]等研究表明月季茎段的最适生根培养基为1/2MS+活性炭1.0g/L,生根率为92.9%。王艳红[9]等以丰花月季叶片、叶柄和茎段为实验材料,结果表明叶片和茎段是诱导愈伤组织的理想外植体。叶贻勋[10]等以月季良种红衣主教带腋芽茎段为外植体进行离体快繁研究表明:在MS+6-BA 2.0mg/L+NAA 0.1mg/L+GA3 1.0mg/L培养基上进行继代增殖,长成的芽苗比较粗壮。 (责任编辑:qin) |
