大豆microRNA调控网络数据库的构建与分析

摘要:microRNA 是一类长度在22nt 左右的非编码小RNA,通过转录后对靶基因的定向剪切,在植物的生长发育和抗病抗逆中都起到重要的调控作用。大豆作为一类重要的蛋白和油料资源,是一种重要的经济作物,因此,研究大豆microRNA对大豆生长发育及抗病抗逆机制研究具有重要意义。本研究收集各大数据库已报道的大量microRNA和降解组测序数据,以此为基础数据构建基于降解组验证的大豆microRNA调控网络数据库,对其基本表进行差、交、并的运算后,构建保守家族调控调控网络,具有组织特异性microRNA调控网络和家族互作网络,讨论和分析microRNA及其靶基因间相互关系的共性、在不同组织中的特性和其在大豆生长发育中作用。41909 毕业论文关键词:microRNA;大豆;降解组测序验证;调控网络;生物信息学;靶基因 Construction and analysis of microRNA regulatory network database in soybean Abstract: microRNAs, a group of 22 nt noncoding small RNAs, play an important role in plant growth and stress response by the cleavage of their target gene during the process of post-transportation. Soybean is considered as vital resource of protein and oil, so it is important to study the microRNA in soybean and the relations between them and their target genes. In this research, we make use of the existent sequencing data and annotation information of microRNAs from various databases to predict the potential cleavage relations. Then we verify these predicted interactions based on degradome libraries we in five different tissues we collected from NCBI. With the degradome-based MIR-TAR information, we construct microRNA regulatory networks and analyze the universal function and specific regulations in these tissues. With bioinformatics and abundant of annotations from various databases, such as miRBase, SoyFN, GENEVESTIGATOR, NCBI, we do the further study about the function of these target protein encoding genes and the pathway involved with microRNA in biological processes of soybean. This research is mainly completed online with abundant reported experiment and sequencing data from reported databases, we provide a new method to learn how to find valuable microRNAs with vital regulatory function in soybean.
