ZFAND5基因在徐淮山羊胎羊组织中表达差异性研究

摘要本次实验主要采集了徐淮山羊胎羊的8种组织,即心、肝、脾、肺、肾、肠、背肌和腿肌,然后采用实时荧光定量PCR的方法,研究分析这8种组织中ZFAND5基因的表达差异性。首先是从徐淮山羊胎羊的8种组织中提取出总RNA,其次反转录成cDNA,再运用real-time PCR技术在各组织中检测ZFAND5基因的表达水平。通过实验得到了在徐淮山羊胎羊的8种组织中ZFAND5基因表达水平依次为心肌(1.0000)、腿肌(0.7318)、背肌(0.1467)、肺(0.0559)、肝(0.0325)、肾(0.0215)、脾(0.0095)、肠(0.0091)。由此可见,ZFAND5基因在徐淮山羊胎羊的肌肉组织中呈高水平的表达,而在其他组织表达量较低,可以推测ZFAND5基因可能与徐淮山羊胎羊的肌肉组织的生长发育和功能有关系。此外,本次实验所得的ZFAND5基因在徐淮山羊胎羊的8种组织中的表达情况,也可为今后研究山羊的ZFAND5基因提供参考。42694 关键词:徐淮山羊 ZFAND5基因 实时荧光定量PCR 差异性表达 The Study on Difference Expression of ZFAND5 Gene in Xuhuai Fetal Goat Organs Abstract The experiments were collected from the eight organizations of Xuhuai fetal goat, namely heart, liver, spleen, lung, kidney, intestine, back muscle and leg muscle, and apply real-time PCR method to study the difference expression of ZFAND5 gene in the eight organizations. The first is extracted the total RNA from the eight organizations of Xuhuai fetal goat , followed by reverse transcribed into cDNA, then using real-time PCR technology to detect the expression levels of ZFAND5 gene in tissues. Through the experiments, the expression levels of ZFAND5 gene in the eight organizations of Xuhuai fetal goat were myocardium (1.0000), leg muscle (0.7318), back muscle (0.1467), lung (0.0559), liver (0.0325), kidney ( 0.0215), spleen (0.0095), intestine (0.0091). Thus, high levels of ZFAND5 gene was expressed in the muscle tissue of Xuhuai fetal goat,while lower expression in other tissues,presumably ZFAND5 gene may be associated with the growth and development and the function of muscle tissue in Xuhuai fetal goat. In addition, the results of this experiment about the difference expression of ZFAND5 gene in the eight organizations also can provide reference for future research on goats ZFAND5 gene.
