小鼠脑瘤模型的构建

摘要脑瘤或颅内肿瘤是指脑内异常细胞的形成。同时也是一种常见病且严重威胁着人类的生命,是患者的死亡率较高的一种疾病。近几年来,肿瘤化疗取得了相当的进步,肿瘤患者生存时间明显延长,然而化疗在杀伤肿瘤细胞的同时,也造成机体骨髓抑制和免疫功能低下,毒副作用大。因此为了更好的研究新药及更好的了解脑瘤的生物学特性,创制出了各类肿瘤的筛选模型。结合比较近年来国内外在此方面的实验研究,构建了本次实验的基础。43939

目的 探讨人脑胶质瘤细胞裸鼠动物模型的建立。方法 借助动物活体成像仪的帮助记录,采用脑立体定位仪微注射方法将体外培养人脑胶质瘤细胞U87MG-LUC(悬浮于含有10%胎牛血清、1%双抗和1%谷氨酸的DMEM培养液中,细胞数分别为5x105和1x106接种于裸鼠左脑前额皮层和第三脑室,接种后观察肿瘤的生长状态。 结果 5x105细胞接种于前额皮层约17天后出现肿瘤四只中两只最后成瘤,1x106细胞接种于前额皮层约11天后出现肿瘤三只中三只最后成瘤,5x105细胞接种于第三脑室约6天后出现肿瘤四只中有四只最后成瘤,1x106细胞接种于第三脑室约9天后出现肿瘤四只中有四只最后成瘤。 结论 本实验的人脑胶质瘤细胞U87MG-LUC用于裸鼠动物模型的建立成瘤率近100%,成瘤时间较好且未见脑外转移。

Abstract Brain tumor or intracranial tumor refers to the formation of abnormal cells in the brain . But also a common and serious threat to human life , is a high mortality rate in patients with a disease . In recent years, chemotherapy has made considerable progress in cancer patients was significantly longer survival time , however, chemotherapy in killing tumor cells , but also cause the body's bone marrow suppression and immune dysfunction, toxic side effects . Therefore, in order to better study of new drugs and a better understanding of the biological characteristics of brain tumors , created out of a variety of tumor screening model . In recent years, combined with comparative experimental researches in this area , to build a foundation for this experiment .

Objective To explore the establishment of human glioma cells in nude mice model.Method with the help of recording animal in vivo imaging instrument , using stereotaxic instrument microinjection methods in cultured human glioma cells U87MG-LUC ( suspension containing 10 % fetal bovine serum , 1% and 1%, double anti- glutamic acid the DMEM culture medium , cells were seeded in nude 5x105 and 1x106 prefrontal cortex and left hemisphere of the third ventricle , tumor growth was observed after vaccination status .The results 5x105 cells were seeded in the prefrontal cortex of about 17 days after tumor four in the last two tumorigenic , 1x106 cells were seeded in the prefrontal cortex of about 11 days after tumor three in the last three tumor , 5x105 cells were seeded in the third ventricle of about 6 four days after the tumor has four final tumorigenic , 1x106 cells were seeded in about nine days after the third ventricle tumor four in the last four tumor .Conclusion The human glioma cell U87MG-LUC used to establish the rate of tumor formation in nude mice in animal models of nearly 100% , and no better time to tumor outside the brain metastases.
