高粱Trihelix转录因子的分子特征与表达分析

摘要:Trihelix转录因子家族在植物生长发育及抵御各种逆境胁迫中发挥重要作用,因其在DNA结构域含有3个串联的螺旋结构而得名,但目前基于高粱全基因组水平鉴定和分析该基因家族的研究并未见相关报道。本文利用生物信息学的方法鉴定出在高粱中含有15个该家族基因,且对其序列特征、基因结构、共线性、染色体与亚细胞定位、表达特征的研究进行了详细综述。结果表明,Trihelix转录因子在高粱的花药、早期花序、25DAP胚芽、25DAP胚乳、20天叶片A、20天叶片B、雌蕊、10DAP种子、5DAP种子均有表达,但每个基因的表达水平有所差异。同时,研究发现高粱的Trihelix转录因子具有很好的共线性,保守性较好,并且在染色体上分布均匀,很少有聚集现象。高粱中的大多数Trihelix转录因子内含子数目较多,表明该家族基因可能在高粱中发生了进化。同时研究表明绝大多数高粱Trihelix转录因子定位到亚细胞中。48623 毕业论文关键词:高粱 ;Trihelix转录因子; 生物信息学 Molecular Characterization and Expression Pattern of the Trihelix Transcription Factor in Sorghum bicolor Abstract: Trihelix transcription factors play important roles in plant growth and stress resistance. Trihelix transcription factors were named because of their DNA structure domain containing three tandem helical structures. In addition, there is no report about sorghum trihelix family genes. In this paper, a total of 15 trihelix genes were identified by using bioinformatics methods, and we further investigate sequence features, gene structure, collinearity, chromosome position, subcellular localization, expression and evolutionary pattern in detail. The results showed that the early Trihelix transcription factors in the anther of sorghum, inflorescence, 25 dap germ, 25 dap endosperm, 20 days of blade, pistil, 10, 20 days of leaf B dap seeds, 5 dap seeds are expressed, but there is obvious difference between the expression level of each gene. At the same time, the study found that Trihelix transcription factor has the very good collinearity, conservative, and distributed evenly on the chromosomes in the sorghum. Most of the sorghum Trihelix transcription factor introns number is more, showing that the pergence between SbTH homologous genes may happen in sorghum. In this paper, the preliminary research provides us for the evolution characteristics of sorghum Trihelix transcription factors, chromosome distribution, expression and subcellular localization. This is a basis of further functionally studying trihelix transcription factors in plant species.
