栀子蓝色素发酵工艺条件研究

摘要: 栀子蓝色素是以茜草科植物栀的果实为原料,利用现代技术提取而成的良好的蓝色着色剂。本实验采用发酵方式从栀子果实中提取黄色素,利用枯草芽孢杆菌将栀子果实中的栀子黄色素转化为栀子蓝色素,并研究其生产工艺条件,通过不同温度、时间、固液比、稳定性等条件探寻最佳提取方式。经实验可知,栀子黄色素在440nm处有最大吸收峰,且最佳提取温度为60℃、提取时间间隔为30min,提取次数为2次,最佳固液比为1:30,最佳溶剂为水,且对栀子果实经行去壳预处理会提高栀子黄色素的提取率。栀子蓝色素的最大吸收峰值在640nm处。培养基的碳源为葡萄糖3%,蛋白胨种类为鱼蛋白胨2%,接种枯草芽孢杆菌培养3d,培养温度为35℃,摇床转速250r/min较好。48960 毕业论文关键词: 栀子黄色素;发酵;栀子蓝色素;工艺条件 Research on fermentation process conditions of gardenia blue pigment Abstract:Gardenia blue pigment is a good colorant,of which raw material is rubiaceae plant gardenia.It 's extracted by modern technology. In this study,the yellow pigment extracted from gardenia fruit by way of fermentation,using Bacillus subtilis transform Gardenia yellow pigment into blue.Research on fermentation process conditions of gardenia blue pigment by different temperature, solid-liquid ratio and extraction times, stability, etc,to explore the best way to extract.The experiment shows that,gardenia yellow pigment has a maximum absorption peak at 440nm.the best extraction temperature of 60 ℃,The best extracting time interval for 30 min,the best solid-liquid extraction times for twice. the best solid-liquid ratio of 1:30,and the best solvent for water,these conditions were the best conditions of the experiment.The pretreatment of Gardenia fruit such as being mechanical shelled will increase the rate of extraction of gardenia yellow pigment.The maximum absorption peak of gardenia blue pigment is at 640nm.Medium comprises carbon source,glucose 3%, nitrogen source , fish peptone2%,Bacillus subtilis,which is cultured 3 days,and were incubated in 250 rpm / min shakers.
