生物絮凝剂高产菌株的筛选及其分子鉴定

摘要[目的]从土壤样品中分离并筛选出具有絮凝活性的菌株并对其进行分子鉴定。[方法]经过平板涂布培养、四区划线纯化和摇床培养后,选取单菌落菌株测定其絮凝活性,然后从中挑取絮凝效果好的菌株,以细菌的 16S rRNA 通用引物对其进行PCR 扩增,根据PCR 产物的电泳检测结果,将其送去测序公司测序,通过比对 NCBI 数据库,从而获得菌株的属名,然后进行同源性比对。[结果]从土壤样品中分离出了一株具有较好絮凝活性的菌株,命名为 BWL-903,测序发现为类芽孢杆菌属,其同源性比对结果表明,菌株 BWL-903 与 Paenibacillusamylolyticus 具有 98.54%的同源性。该论文有图 6幅,表 1个,参考文献 28篇。51089 毕业论文关键词:微生物絮凝剂 分子鉴定 16S rRNA Screening of High Yield Strain of Bio Flocculant andIts Molecular Identification Abstract [Objective]The strains with flocculating activity were isolated and screened fromsoil samples and their molecular identification was carried out.[Method]After coatedplate culturing, purification crossed and shaking culture, single colonies wereselectedto test the flocculating activity and then the strain which has a fineflocculating activity were selected, PCR amplification of the 16S rDNA was identifiedby Tiangen Biotechnology, and compared to the sequences available in the GenBankfrom the National Center for Biotechnology Information (NCBI) Database,and thenthe homology comparison was carried out.[Result]A strain , which has a fineflocculating activity was isolated from soil samples, named BWL-903, which wasidentified as Bacillus.And its homologous alignment results showed that, the strainBWL-903 and Paenibacillus amylolyticus with 98.54% homology.
