水稻ABC1激酶ABC1K3的突变体分析

摘 要:ABC1(Activity of bc1 complex)家族是一个大的、功能多样的蛋白质家族,其成员普遍存在于原核和真核生物中。水稻基因组含有 16 个推定的 ABC1 激酶基因,但其功能目前尚不明确。本研究利用 T-DNA 插入突变体对水稻 ABC1 激酶 ABC1K3 的生理和分子功能进行了鉴定。结果表明,osabc1k3纯合突变体的株高、育性和叶片叶绿素含量都显著下降,并且在强光处理下出现了严重的黄化和枯死表型,这说明osabc1k3突变体植株对强光处理更加敏感。Real-time PCR结果表明,osabc1k3基因突变影响了叶绿体异戊二烯酯合成相关基因的表达。这些结果表明,OsABC1K3激酶可能在叶绿体异戊二烯酯合成和强光胁迫应答中发挥重要作用。55870 毕业论文关 键 词:水稻,ABC1基因,T-DNA插入突变体,强光胁迫,异戊二烯酯合成Abstract: ABC1 (activity of bc1 complex) family is a large and functionally perse protein family, whose members are widely found in prokaryotes and eukaryotes. The rice genome contains 16 putative ABC1 kinase genes, but their function remains illusive. In this study, the OsABC1K3 kinase was characterized using T-DNA insertion mutants. The osabc1k3 mutants harbored decreased plant height, fertility and leaf chlorophyll contents and exhibited severe degreening and necrosis under high light treatment. Real-time PCR analysis showed that osabc1k3 mutation affected the expression of chloroplast prenyllipid biosynthetic pathway genes. These results suggested that OsABC1K3 might play essential roles in chloroplast prenyllipid biosynthesis and high light stress response.
