女贞子果不同部位齐墩果酸和熊果酸含量测定及其提取工艺

摘要:本文研究从苏北地产女贞子中提取齐墩果酸和熊果酸的适宜于工业生产的方法。实验方法为将女贞子果分成果肉、果壳、果仁三个部分,分别将各个部位用加热和冷冻两种方式干燥且分解成八种原料后,用回流提取法和超声提取法在不同浓度的乙醇与不同质量百分比浓度的氢氧化钠溶液的混合溶剂中提取,过滤,并定容后,采用高效液相色谱法进行含量测定分析。结果表明:加热干燥与冷冻干燥方式没有区别;在相同提取条件下,齐墩果酸和熊果酸在不同部位的含量分别为果肉(25.5920±0.0072, 6.7675±0.0040mg/g)>果子(5.5828±0.0016,2.5319±0.0241mg/g)>果壳(4.4005±0.1505,2.1321±0.1746mg/g)>果仁(0.3998±0.0133, 0.1578±0.0129 mg/g)。以果肉为原料,经干燥后,用质量百分比浓度为5%的NaOH水溶液与无水乙醇配制成的95%乙醇的混合溶剂,按料液比1:9混合后,在超声下进行提取为最佳提取工艺。得到的齐墩果酸、熊果酸含量分别为29.5521±0.0180mg/g、13.1175±0.0618mg/g。55993 毕业论文关键词:齐墩果酸,熊果酸,超声提取法,高效液相色谱法 Abstract: In this paper, a method for the suitable industrial production extracted oleanolic acid and ursolic acid from fruit of Ligustri Lucidum from Subei in Jiangsu province were researched. Experimental methods were that the fruit of Ligustri Lucidum was shucked into pulp, shell and pip by manual work, and each fraction was dried by both in heating and freezing drying and pided down into eight kinds of raw materials respectively, and then they were extracted by using reflux and ultrasonic method with different concentrations of ethanol and different mass percent concentration of sodium hydroxide solution of mixed solvent. After mixture were filtered and solution were constant volume, oleanolic acid and ursolic acid were determined by using high performance liquid chromatography (HPLC) method. The results show there is no difference for two compounds by heating dry and freeze dry way; In the same extraction condition, the test results as follows: pulp (oleanolic acid 25.5920 ± 0.0072 mg/g, ursolic acid 6.7675 ±0.0040 mg/g) > fruit (oleanolic acid 5.5828 ±0.0016 mg/g, ursolic acid 2.5319 ±0.0241 mg/g) > shells (oleanolic acid 4.4005 ± 0.1505 mg/g, ursolic acid 2.1321 ± 0.1746 mg/g) > pip (oleanolic acid 0.3998± 0.0133 mg/g, ursolic acid 0.1578 ±0.0129 mg/g). Pulp of the fruit of Ligustri Lucidum is batter crude to extract oleanolic acid and ursolic acid and extract technology was that pulp drying was immersed with the mass percentage concentration of 5% NaOH aqueous solution and mixture of anhydrous ethanol 95% ethanol mixed solvent, mixes the material liquid than 1:9, under the ultrasonic extraction as the best extraction process.
