不同的氮源对里氏木霉产纤维素酶的影响的研究

摘要:  本论文以里氏木霉Rut C-30(Trichoderma reesei)为产酶菌种,研究了不同氮源对里氏木霉产纤维素酶的影响。分别在固定硫酸铵含量1.4g/L或5g/L 的基本培养基分调节玉米浆(CSL)的浓度(5、3、2、1g/L)来测定纤维素酶酶活随时间的变化;在含有不同浓度的硫酸铵的基本培养基测定酶活随时间的变化(5、3、2、1.4g/L);在含有不同浓度的尿素(0.1%、0.2%、0.3%、0.4%、0.5%)的基本培养基测定酶活随时间的变化。实验表明不同的氮源对里氏木霉产纤维素酶影响不大,其中硫酸铵含量1.4g/L 的基本培养基CSL浓度为1g/L的酶活最高为6.07U/ml;而硫酸铵含量为5g/L的培养基中CSL浓度为2g/L的酶活最高为6.02U/ml;当硫酸铵浓度为3g/L时酶活最高为5.87U/ml;0.3%的尿素能够有效的提高产酶能力。5优尔6

Abstract: This thesis uses Trichoderma reesei Rut C-30 as cellulase production strain to study the effects of different nitrogen sources on the production of cellulase. Basic medium with 1.4g/L or 5g/L ammonium sulphate of corn steep liquor (CSL) concentration (5, 3, 2, 1g /L) to determine the change of enzyme activity over time; Enzyme activity was determined over time in basic medium containing different concentrations of ammonium sulfate (5, 3, 2, 1.4g /L) or different concentrations of urea (0.1%, 0.2%, 0.3% , 0.4%, 0.5%). Experimental results show that the different nitrogen sources do not have much effect on the T. reesei cellulase production. Basic medium with 1.4g/L ammonium sulfate has the highest activity when CSL concentration is 1g/L; While Basic medium with 5g/L ammonium sulfate has the highest activity when CSL concentration is 2g/L; When changing ammonium sulfate concentration, 3g/L ammonium sulfate gives maximum enzyme activity; When changing urea concentration, 0.3% urea gives the highest cellulase activity.
