黄牛bta-miRNA-660遗传多态性与生长性状关系分析

摘要秦川牛的bta-miRNA-660是一种非编码的miRNA。为了达到研究黄牛bta-miRNA-660遗传多态性的目的,本次实验采用了PCR-SSCP技术以及DNA测序等技术,从而为黄牛优秀生长发育性状的分子育种提供重要依据。可找出相关遗传标记,达到保留优势性状的目的。为秦川牛的高效选育,其分子标记库的建立,种质资源的保护与利用提供遗传学依据。采用PCR-SSCP方法对秦川牛bta-miRNA-660基因进行分析,实验结果清晰显示出bta-miRNA-660基因存在多个遗传多态性,之后通过DNA测序加以证明。其遗传位点表现为AA型、BB型以及AB型三种基因型,其频率分别为0.103,0.308及0.590。A等位基因的基因频率为0.3974,B等位基因的基因频率为0.6026。经与秦川牛生长性状的关联分析发现,不同基因个体之间的生长性状存在明显差异,由此可以证明该位点可以作为秦川牛生长性状选择的分子标记。71677

Cattle bta-miRNA-660 Genetic Polymorphisms with Growth Analysis

Abstract Qinchuan cattle’s bta-miRNA-660 is a non-coding miRNA. The study used PCR-SSCP technique and DNA sequencing technology to analyze the genetic polymorphism bta-miRNA-660 gene in Qinchuan cattle in our country, thus providing an important basis for the molecular breeding cattle excellent growth traits. In order to identify genetic markers and economic traits most relevant. Qinchuan cattle breeding for the efficient establishment of the molecular tag libraries, protection and utilization of germplasm resources to provide genetic basis. Using PCR-SSCP method Qinchuan bta-miRNA-660 gene analysis showed that bta-miRNA-660 gene is present more songs genetic polymorphism, and later the same may be proved through DNA sequencing. After genetic loci showed AA genotype, BB, and type AB type three genotypes which frequencies were 0.103,0.308 and 0.590. A allele frequency of 0.3974, B allele frequency of 0.6026.After Qinchuan and growth traits association analysis found that different inpiduals have significant growth traits genetic differences, indicating that the site can be used as growth traits in Qinchuan Cattle selection marker.
