辣椒叶提取物的抗肿瘤活性检测

摘要:背景:由于癌症化疗给病人带来的副作用,具有肿瘤预防和治疗的食品收到受到国内外学者的关注。目的:从食疗角度研讨辣椒叶提取物对肿瘤的预防和治疗作用。方法:采用Alamar blue法检测辣椒叶提取物对肝癌细胞HepG2细胞活力的影响; Hoechst33258荧光染色检测辣椒叶提取物对HepG2细胞的形态的改变。AnnexinV/PI试剂盒检测辣椒叶提取物是否促进肝癌细胞的凋亡;Caspase3活性检测试剂盒研究辣椒叶提取物对Caspase3活性的调节。结果:辣椒叶提取物能够剂量依赖性的抑制HepG2细胞的增殖活性;倒置显微镜和荧光显微镜观察发现,辣椒叶提取物能够改变肝癌细胞HepG2的细胞形态,导致细胞死亡;凋亡试剂盒检测结果证实辣椒叶提取物能够剂量依赖性的诱导HepG2细胞凋亡;辣椒叶提取物处理过的HepG2细胞中Caspase3的活性明显增加。结论:辣椒叶提取物是通过诱导HepG2细胞的凋亡来实现其抗肿瘤活性,该研究为进一步研究其分子机制提供实验依据。76471

The antitumor activity detection of hot pepper leaf extraction

Abstract: Background Because of the severe side-effects of radiotherapy and chemotherapy for cancer, more and more researchers pay attention to the exploitation of anti-cancer plantal foods。 Objective To study the effect of pepper leaf extraction on the proliferration of cancer cells and the morphological detection of their apoptosis。 Methods The proliferation of HepG2 cells was detected by Alamar Blue assay;Hoechst33258 staining was used to detecte the apoptosis of HepG2 cells。 AnnexinV/PI double staining was used to detect the apoptotic rate of HepG2 cells induced by pepper leaf。 The activity of Caspase3 was detected by Caspase 3 kit。 Results The pepper leaf extraction inhibited the proliferation of HepG2 cells in a dose-dependent manner。 The morphology of HepG2 cells treated with pepper leaf extraction was changed and the cells wad dead。 The apoptotic rate of HepG2 cells treated with pepper leaf extraction was increased in a dose-dependent manner。 And pepper leaf extraction can significantly increase the Caspase3 activity in HepG2 cells。 Conclusion The pepper leaf extraction inhibited the proliferation of HepG2 cells by inducing their apoptosis。 This study provides experimental basis for further investigation of the molecular mechanism of pepper leaf extraction anti-tumor activity。
