轮枝镰刀菌中Swi4-Swi6复合体生物学功能研究

摘要:轮枝镰孢菌所造成的的玉米穗腐和茎基腐病是玉米上重要的一种病害。通过对轮枝镰孢菌突变体∆FvSwi4和∆FvSwi6对玉米粒和玉米茎干的致病性检测发现,在玉米粒上突变体∆FvSwi4和∆FvSwi6的菌丝明显减少,玉米粒的腐烂现象明显减弱。在玉米茎干上,发现突变体∆FvSwi4和∆FvSwi6的褐色病斑长度明显小于野生型菌株Fv102。这表明突变体∆FvSwi4和∆FvSwi6对玉米致病性减弱。77898

真菌的无性生殖对真菌在环境中存活,并对真菌对对植物侵染是保证迅速扩展与是植物发病有重要意义。在不同培养基上的生长速率说明,突变体∆FvSwi4和∆FvSwi6在PDA、CM和MM培养基上的生长速率菌小于野生型菌Fv102。对分生孢子的产孢量的测定表明,这两个基因会影响轮枝镰孢菌的分生孢子的形成,从而导致轮枝镰孢菌的生长缓慢。有性生殖是真菌进化中必不可少的阶段,对轮枝镰孢菌的子囊壳产生的实验表明,突变体∆FvSwi4和∆FvSwi6完全无法产生子囊壳,说明这两个基因对轮枝镰孢菌的有性生殖过程也有着重要影响。

敲除突变体∆FvSwi4、∆FvSwi6影响真菌色素的合成,本研究进一步扩大培养,通过硅胶层析、凝胶、HPLC等分离技术分离提纯色素化合物,并通过核磁共振分析、质谱分析等技术分析其化学结构,一共得到3个化合物。

Study on the Biological Function of Swi4-Swi6 Complex in FUMARIUM VERTICILLIOIDES

Abstract:Maize ear rot and stalk rot caused by Fusarium verticillioides is an important disease on maize。 The pathogenicity of FvSwi4 and FvSwi6 to Fusarium verticillioides and corn stalk was detected by Fusarium verticillioides 。 The FvSwi4 and FvSwi6 of the mutants in maize grains were significantly reduced, and the decay of maize grains was significantly weakened。 In the corn stem, the mutant Delta FvSwi4 and delta FvSwi6 Brown lesion length was significantly less than the 

wild type strain Fv102。 These results indicated that the mutant FvSwi4 and delta FvSwi6 reduced pathogenicity of maize。

The asexual reproduction of fungi can survive in the environment, and it is important to ensure the rapid expansion and the occurrence of plants。 The growth rate in different media showed that the growth rate of mutant FvSwi4 and FvSwi6 on PDA, CM and MM medium was lower than that of wild type Fv102。 The conidial production of conidia showed that these two genes could affect the formation of conidia of Fusarium verticillioides, which led to the slow growth of Fusarium verticillioides。 Sexual reproduction is essential in fungal evolution stage, that of perithecia Fusarium verticillioides production experiments, mutant FvSwi4 and FvSwi6 cannot produce perithecia, explain the process of sexual reproduction of these two genes to Fusarium verticillioides has important effect。

The knockout mutant of delta FvSwi4, delta FvSwi6 affect the synthesis of fungal pigments, this study further expand the culture, purification of pigment compounds separated by silica gel chromatography, gel and HPLC separation technology, analysis of its chemical structure and by NMR analysis, mass spectrum analysis technology, a total of 3 compounds。
