摘要:亚硝酸盐广泛存在于食品、土壤及水中,因为亚硝酸盐具有良好的抑菌作用,而被允许用作食品添加剂起到防腐作用;又因其具有良好的发色作用,常常被用在肉类制品中。亚硝酸盐是一种氧化剂,可致使细胞失去与氧结合的能力,并且是潜在的致癌物质。因此亚硝酸盐超标问题近来引起人们极大的关注。植物乳杆菌是一种益生菌,研究发现,植物乳杆菌可以产生降解亚硝酸盐的酶,即亚硝酸还原酶;相较之下更安全高效的方法是生物酶法,这也是一种新的降解亚硝酸盐的方法。本课题利用实验室保藏的植物乳杆菌菌种,从中得到表达亚硝酸还原酶的基因片段,先将目的基因进行TA克隆并转化到E.coli DH5α中,经蓝白斑筛选后,将所得质粒双酶切,得到可用的目的基因片段。再构建pET-21a(+)-NiRs质粒并转入E.coli BL21(DE3)中。本研究课题得到的重组质粒可用于对植物乳杆菌中亚硝酸还原酶基因的表达的研究。20040

Cloning of Lactobacillus plantarum nitrite
reductase gene
Abstract: Nitrite is widely present in food, soil and water. Because the nitrite has good bacteriostatic effect, it is permitted as a food additive plays antiseptic effect. And because of its good hair color, nitrite is often used in meat products. Nitrite is an oxidizing agent, it may cause the cells lose the ability of combining with oxygen, and is a potential carcinogen. Therefore, the nitrite content exceed the standard problem has aroused great attention. L. Plantarum is the human probiotic that can produce nitrite reductase. Enzymatic degradation of nitrite is a safe and effective new way. In this subject, we use Lactobacillus plantarum strains that preserved in our laboratory to get nitrite gene sequence. We use TA cloning firstly, and transform it into E.coli DH5α. Through blue-white spot screening and double digests, target fragment would be available. Then construct plasmid pET-21a(+)-NiRs and transform it into E.coli BL21(DE3). The product we obtain could be used in the study of nitrite reductase gene of Lactobacillus plantarum.
