摘要核糖体(ribosome)作为细胞内负责蛋白质合成的重要细胞器一直为人们所熟知。早期的研究认为核糖体蛋白仅有组成核糖体的功能,近年来,有越来越多的核糖体蛋白基因核外生物学功能的报道,最新的研究表明RPL10A参与了植物对DNA病毒的免疫防御。
拟南芥(Arabidopsis thaliana)是研究植物与病原生物的互作、剖析植物抗病机制的最重要的模式植物之一。值得一提的是,拟南芥有三个RPL10同源基因,分别被命名为AtRPL10A,AtRPL10B和AtRPL10C。前期实验证明PVX/AtRPL10A重组RNA接种本氏烟草后,植株出现矮化及病斑加重的现象,同时还对其育性产生了严重的影响。在此基础上,本课题采用病毒诱导基因沉默技术(VIGS),在本氏烟草系统中研究拟南芥RPL10A同源基因RPL10B的功能。87965
实验结果表明,接种PVX/AtRPL10B重组病毒载体后的本氏烟草也出现了矮化现象,此外植株叶片也有较严重的卷曲变形和一定的褪绿现象。
Abstract Ribosome has been well-known as one of important organelles responsible for protein synthesis in cells。 Early studies suggested that ribosomal protein genes only function in setting up ribosomes。 Recently, more and more extra-ribosome functions of ribosomal protein genes have been reported, and recent studies have shown that RPL10A is involved in a plant antiviral immunity mechanism。
Arabidopsis thaliana is one of the most important model plant for studying the interaction between plants and pathogens, and analyzing the mechanism of plant antiviral immunity。 It is worth to be mentioned that Arabidopsis thaliana has three RPL10 homologous genes: AtRPL10A, AtRPL10B and AtRPL10C。 Previous experiments have showed that after being inoculated PVX-/AtRPL10A recombinant virus, the plants of Nicotiana benthamiana(N。B。) were not only dwarf and severe lesion increased, but also had a serious impact on its fertility。 On this basis, the function of Arabidopsis thaliana RPL10B was studied in the PVX-N。B。’s system by virus-induced gene silencing (VIGS)。
The results showed that the plant N。B。 was also dwarfed after inoculating PVX / AtRPL10B recombinant virus vector。 In addition, the leaves of the plant had severe leaf distortion and moderate chlorosis。
