铁皮石斛组织培养的优化及仿野生栽培

摘要本研究是以铁皮石斛(Dendrobium officinale Kimura et Migo)试管苗为材料，进行快速繁殖的优化研究，主要步骤包括丛生芽的增殖、苗的生根、炼苗、移栽以及仿野生栽培。通过株数、株高、根数、根长的测定及成活率的统计分析，探究其较优的生长条件。研究结果表明：1/2MS+0。1 mg•L-1 NAA +0。1 mg·L-1 PP333 培养基在增粗幼苗茎杆及增加其生根率方面有着显著优势。炼苗两天的石斛苗在之后的实验中成活率最高。移栽后，栽培在在树皮中的石斛苗生长状况最好。此外，每年四月份较五月份、八月份和九月份更适合石斛苗的生长。仿野生环境中，最适宜幼苗生长的环境条件为于水杉树上贴树生长。88030

This experiment was based on studying rapid propagation of Dendrobium officinale Kimura et Migo in vitro,mainly including adventitious bud multiplication and rooting, hardening, transplanting and the imitation of the wild cultivation。 Through the statistics of stem height,root length,the number of stems and roots, and the calculation of the survival rate , to explore the optimal growth conditions。 The result shows that culture medium (1/2MS+0。1 mg• L-1 NAA +0。1 mg• L-1 PP333) has a significant advantage in enlarging seedling stem and increasing rooting rate。 Seedlings which were exercised for two days had the highest survival rate of experiment。 After transplanting,seedlings of Dendrobium candidum grow best in the bark。 In addition, April is the more suitable month for the growth of seedlings than May,August and September。In the imitation of the wild cultivation, dawn redwood is more appropriate for growing seedlings。
