摘 要动物大脑是调控身体机能和对外界信息进行处理的中央枢纽,大脑对信息的处理涉及的生物学进程非常复杂。视觉是脊椎动物接收外界信息的主要方式,大脑视觉区域接受视网膜神经节细胞的信息传入后进行加工处理。而这个过程需要蛋白的合成和调控。我们使用爪蟾作为模式动物,研究视觉刺激引起的蛋白合成变化。本项目对47期蝌蚪进行4小时视觉刺激,前期实验结果表明,视觉刺激会引起蝌蚪视觉逃避行为的变化,逃避率明显增加,并且发现这种变化和大脑内组蛋白乙酰化水平有关。为了验证这种变化是否与大脑内蛋白合成有关。我们提取视觉刺激后的蝌蚪视顶盖,进行western blot 检测,发现视觉刺激以后,I类组蛋白去乙酰化酶HDAC1、HDAC2,HDAC3蛋白合成都显著性增加。说明视觉刺激的过程会引起视顶盖中的HDAC1、HDAC2,HDAC3表达发生变化,这些蛋白参与了大脑对视觉刺激信息处理的过程,在调控蝌蚪逃避行为过程中起了重要作用。89949
Abstract Animal's brain is known as a central system to regulate the body function and information processing。 It is very complicated because the information processing involves many biological processes。 Receiving the visual information is the main pathway for vertebrates to communicate with the world。 The visual information from retinal ganglion cells is processed in the visual regions of brain, which requires protein synthesis and regulation。 We took Xenopus laevis as a model animal to measure the changes of protein synthesis by visual stimulation。 The tadpoles at stage 47 were exposed to visual stimulation for 4 hours。 Our preliminary experimental results showed that the avoidance index was significantly increased by visual stimulation。 We found that the change was correlated with the level of histone acetylation。 In order to test whether this change was associated with protein synthesis in the optic tectum, we extract the proteins of the optic tectum after visual stimulation for Western blot。 We found that visual stimulation caused a significant increase in protein synthesis of class I histone deacetylases, including HDAC1, HDAC2 and HDAC3。 Our data suggested that visual stimulation can cause changes of expression of HDAC1, HDAC2 and HDAC3 in the optic tectum of Xenopus laevis, which were involved in the information processing and escape behavior。
