摘    要 目的 繁殖和鉴定Isl2/LacZ双基因同源基因重组。方法：将Isl2基因与LacZ基因通过同源基因重组的方式培养出的质粒注射到受精卵中，将胚胎移植到代孕母鼠体内，经过培育得到杂合子与纯合子小鼠进行饲养和繁殖，提取母体中胚胎期小鼠，取其尾部的DNA，用PCR方法扩增小鼠DNA在通过检测子代小鼠基因型，通过对LacZ染色的结果分析可以分析Isl2的分布与含量，从而进行进一步对Isl2基因在体内的作用，当今研究中，该基因可在大脑，脊髓，视网膜，耳蜗里面有表达，如果敲除该基因会导致这些位置在胚胎发育期出现畸形，如果将这个基因在全身敲除，小鼠胚胎会在出生第一天死亡。该基因是否表达和表达量的研究有助于我们了解试听等方面的病因与缺陷。90159
