华鳈sox9基因克隆与多态性分析

摘要：Sox 基因家族是在 HMG 保守盒内与哺乳动物性别决定基因 SRY 的相似性大于 60% 的一系列基因，并且 Sox 基因紧靠对性别决定和分化具有决定作用的 SRY 基因的下游。而 Sox9 基因是 Sox 基因家族中较为保守的基因。华鳈（Sarcocheilichthys sinensis）属鲤形目，鲤科，鳈属，在我国平原地区的江河、湖泊均有分布。本研究利用洪泽湖中华鳈作为研究对象，采用 RT-PCR 法克隆获取了华鳈的 Sox9 基因序列，并进行了华鳈 Sox9 基因与近缘物种的同源性和进化分析。结果获得华鳈 Sox9 基因的 cDNA 片段长度为 1377 bp，编码 379 个氨基酸。本研究获得的数据将为华鳈中 Sox9 基因功能分析，性别决定机制研究，基因多态位点与生长性状的关联分析以及今后开展华鳈的分子标记辅助育种均具有参考意义。85857

Abstract ： The Sox gene family is a series of genes which have more than 60% sequence similarity with the mammalian sex determining gene SRY in their conserved HMG boxes, and they locate at the downstream of SRY gene。 Sox9 gene is a conserved gene in the Sox family with functions in sex determination and differentiation。 Sarcocheilichthys sinensis which belongs to the genus Sarcocheilichthys in subfamily Gobioninae of the Cyprinidae family mainly distributes in rivers and lakes in plain areas of China。 In this research S。 sinensis from the Hongze Lake was applied to clone Sox9 gene using RT-PCR technology and homologous and evolutionary analyses were also performed。 Results showed that a cDNA fragment of Sox9 gene was obtained, the length of which was 1377 bp encoding a peptide with 379 amino acids。 The data got in this study will be helpful for future studies on functional analyses of Sox 9, sex determination investigation, association analysis between polymorphic sites and economic traits and marker assisted selection breeding in this fish。
