摘要:目的 本研究拟通过用红豆杉树皮作为木蹄层孔菌的培养基,通过检测相关化合物的情况来反应木蹄层孔菌与红豆杉树皮相互作用后两者的化学成分变化,为红豆杉皮培养木蹄层孔菌的相关基础研究提供基础。方法 以红豆杉皮为原料,粉碎后平均分配,制作多份培养基,用以培养木蹄层孔菌,然后使用甲醇作为溶剂,和等量未使用红豆杉皮培养的木蹄层孔菌通过回流提取的方法提取有效成分,并分别使在490 nm处通过测定紫外吸收光判断多糖浓度、GC-MS测定木蹄层孔菌中三萜化合物种类以及使用HPLC测定红豆杉皮化合物变化。结果 红豆杉皮培养的木蹄层孔菌中多糖的含量高于对照,HPLC测定后发现红豆杉皮化合物有部分成分变化明显。结论 红豆杉皮中化合物Sinenxans A、Sinenxans B、Sinenxans C培养前后有显著变化,红豆杉中的代谢产物会被木蹄层孔菌利用,树皮中的这三种物质会经过转化,成为中间产物,然后通过中间产物转变成新的物质。49319

Analysis of ingredients content in the process of using the bark of the Taxus chinensis to cultivate Fomes fomentarius

Abstract: Purpose: This study was intended to provide the basis for the study of the related basic research of the Fomes fomentarius by using the bark of the Taxus chinensis as a culture medium for the cultivation of Fomes fomentarius and detecting the related compounds to reflect the changes in the chemical composition of the interaction between the bark of the Taxus chinensis and the Fomes fomentarius.  Methods: Using the bark of the Taxus chinensis as the raw material, then crushing and distributing it into several conical flasks equally to culture Fomes fomentarius. Using the blank Fomes fomentarius (without the bark of the Taxus chinensis) as comparison, both add methanol to conical flasks as solvent, then using reflux extraction method to extract effective component. The concentration of polysaccharide was measured by the use of ultraviolet spectrophotometry in the determination of light absorption at 490 nm, and using GC-MS to analyze polysaccharides of Fomes fomentarius, and using HPLC to analyze triterpene content. Results: The Fomes fomentarius that had been cultured on the bark of the Taxus chinensis has much more polysaccharides than the blank Fomes fomentarius. Compared with the blank Fomes fomentarius, the part of the triterpene content of the Fomes fomentarius that had been cultivated by the bark of the Taxus chinensis has changed obviously. Conclusion: We can find the significant changes of Sinenxans A, Sinenxans B and Sinenxans C after this culture. The metabolites of the bark can be used by the Taxus chinensis. These three kinds of compounds are converted to intermediate products, and then the intermediate products will produce new compounds.
